Abstract

Stapled peptides and methods of using them to inhibit the Kras/Raf interaction are disclosed herein.

What is claimed is: 
     
       1. A compound having the following structure: 
       
         
           
           
               
               
           
         
         wherein X1 and X2 are amino acid residues bonded together. 
       
     
     
       2. The compound of  claim 1 , wherein X1 is an aspartic acid residue. 
     
     
       3. The compound of  claim 1 , wherein X1 is a glutamic acid residue. 
     
     
       4. The compound of  claim 1 , wherein X2 is a lysine residue. 
     
     
       5. The compound of  claim 1 , wherein X1 and X2 are modified amino acid residues that form an alkyl, alkenyl, alkynyl, heteroaryl, aryl, ether, ester, thioether, thioester, amino, or amido containing bridge. 
     
     
       6. The compound of  claim 1 , having the following structure 
       
         
           
           
               
               
           
         
         wherein X 3  is an alkyl, alkenyl, alkynyl, heteroaryl, aryl, ether, ester, thioether, thioester, amino, or amido moiety having from 5 to 25 atoms. 
       
     
     
       7. A method for treating a tumor, comprising the step of administering to a patient in need thereof a therapeutically effective amount of a composition comprising the compound of  claim 1 .
