Abstract

A fusion protein comprising factor VII (FVII) and transferrin according to the present invention has an improved specific activity of FVII compared to existing FVII fusion proteins comprising other fusion partners than transferrin, and thus can be effectively used in a therapy using FVII.

Claims

exact text as granted — not AI-modified
The invention claimed is:

       1. A fusion protein comprising factor VII (FVII) and transferrin,
wherein said transferrin is a wild-type human transferrin linked to the C-terminus of said FVII, which further comprises a linker between the FVII and the transferrin,
wherein the linker is a peptide comprising the amino acid sequence of SEQ ID NO: 12 which comprises a thrombin cleavage site,
wherein the fusion protein has substantially equivalent activity compared to an unfused human FVII,
and wherein a half-life of the fusion protein is about 2˜4 times longer than that of an unfused human FVII.

       2. The fusion protein of claim 1 , wherein said FVII is a polypeptide comprising the amino acid sequence of SEQ ID NO: 1.

       3. The fusion protein of claim 1 , wherein said transferrin is a polypeptide comprising the amino acid sequence of SEQ ID NO: 2.

       4. The fusion protein of claim 1 , wherein one or more amino acids translated from a restriction enzyme recognition site are present at one end or both ends of the linker.

       5. A DNA coding for the fusion protein of claim 1 .

       6. The DNA of claim 5 , wherein the DNA comprises the nucleotide sequence of SEQ ID NO: 23 or 24.

       7. A recombinant vector comprising the DNA of claim 5 .

       8. An isolated host cell comprising the recombinant vector of claim 7 .

       9. The host cell of claim 8 , which is selected from the group consisting of CHO cell, BHK21 cell, HEK293 cell and Hep G2 cell.
