Abstract
The present disclosure provides compositions and methods for the delivery of immune cells to treat un-resectable or non-resected tumor cells and tumor relapse. The compositions comprise (i) a structure comprising an injectable polymer or scaffold comprising pores; (ii) lymphocytes disposed within the structure, (iii) at least one lymphocyte-adhesion moiety associated with the structure; and (iv) at least one lymphocyte-activating moiety associated with the structure, and optionally an immune stimulant.

1. An alginate polymer scaffold coated with a synthetic collagen-mimetic peptide comprising SEQ ID NO: 2, wherein the scaffold houses: porous silica microparticles with a pore diameter of about 100 Å and wherein the microparticles are (a) coated with anti-CD3 antibodies and anti-CD28 antibodies and (b) loaded with interleukin 15.
2. A composition comprising (i) an injectable alginate polymer or an implantable alginate polymer scaffold comprising pores; and (ii) SEQ ID NO: 2, anti-CD3 antibodies, anti-CD28 antibodies, and interleukin 15.
3. The composition of claim 2 , further comprising lymphocytes.
4. The composition of claim 2 , wherein the anti-CD3 antibodies and anti-CD28 antibodies are bound to or incorporated in one or more particles.
5. The composition of claim 2 , further comprising anti-CD137 antibodies.
6. The composition of claim 2 , wherein the alginate polymer is injectable.
7. The composition of claim 2 , wherein the alginate polymer is an implantable scaffold, and the SEQ ID NO: 2 is associated with the scaffold in a bioactive coating on the scaffold.
8. The composition of claim 2 , wherein the alginate polymer is an implantable scaffold, and the anti-CD3 antibodies and anti-CD28 antibodies are associated with particles embedded in the pores of the scaffold.
9. The composition of claim 2 , wherein the alginate polymer is an implantable scaffold and the anti-CD3 antibodies and anti-CD28 antibodies are associated with particles attached to the surface of the scaffold or are embedded in the scaffold.Cited by (0)
