Abstract

The present invention relates to novel antibodies and fragments that bind to a V-domain Ig Suppressor of T cell Activation (VISTA), and methods of making and using same. Methods of use include methods of treatment of cancer, including leukemias, lymphomas, solid tumors and melanomas.

What is claimed is: 
     
         1 . An isolated or recombinant nucleic acid which encodes for an antibody or antibody fragment thereof comprising an antigen binding region that binds to a V-domain Ig Suppressor of T cell Activation (VISTA), which antibody:
 (i) comprises a VH domain comprising a VH CDR1 having the amino acid sequence of SEQ ID NO: 25, a VH CDR2 having the amino acid sequence of SEQ ID NO:26 and a VH CDR3 having the amino acid sequence of SEQ ID NO:27, and   (ii) comprises a VL domain comprising a VL CDR1 having the amino acid sequence of SEQ ID NO:28, a VL CDR2 having the amino acid sequence of SEQ ID NO: 29 and a VL CDR3 having the amino acid sequence of SEQ ID NO: 30.   
     
     
         2 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises antibody or antibody fragment is a Fab, F(ab′)2, or scFv antibody fragment. 
     
     
         3 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment is a human or humanized antibody. 
     
     
         4 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable heavy chain polypeptide having a sequence at least 90% identical to SEQ ID NO:37. 
     
     
         5 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable heavy chain polypeptide having a sequence at least 95% identical to SEQ ID NO:37. 
     
     
         6 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable heavy chain polypeptide identical to SEQ ID NO:37. 
     
     
         7 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide having a sequence at least 90% identical to SEQ ID NO:44. 
     
     
         8 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide having a sequence at least 95% identical to SEQ ID NO:44. 
     
     
         9 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide identical to SEQ ID NO:44. 
     
     
         10 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide having a sequence at least 90% identical to SEQ ID NO:44 and a variable heavy chain polypeptide having a sequence at least 90% identical to SEQ ID NO:37. 
     
     
         11 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide having a sequence at least 95% identical to SEQ ID NO:44 and a variable heavy chain polypeptide having a sequence at least 95% identical to SEQ ID NO:37. 
     
     
         12 . The nucleic acid of  claim 1 , wherein the encoded antibody or antibody fragment comprises a variable light chain polypeptide having a sequence identical to SEQ ID NO:44 and a variable heavy chain polypeptide identical to SEQ ID NO:37. 
     
     
         13 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises a human constant region. 
     
     
         14 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises a human IgG1 constant domain. 
     
     
         15 . The nucleic acid of  claim 11 , wherein the encoded human constant domain comprises a mutation which impairs FcR binding or protease cleavage. 
     
     
         16 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises a human IgG1 heavy constant domain having the human IgG1 heavy constant domain sequence comprised in SEQ ID NO:55, SEQ ID NO:59, SEQ ID NO:60 or SEQ ID NO:61. 
     
     
         17 . The nucleic acid of  claim 1 , which comprises a human IgG1 light constant domain having the human IgG1 light constant domain sequence comprised in SEQ ID NO: 56. 
     
     
         18 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises a human IgG1 light constant domain having the human IgG1 light constant domain sequence comprised in SEQ ID NO:56. 
     
     
         19 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises the heavy chain polypeptide of SEQ ID NO:55, 59, 60 or 61 and the light chain polypeptide of SEQ ID NO: 56. 
     
     
         20 . The nucleic acid of  claim 1 , wherein the encoded antibody comprises the heavy chain polypeptide of SEQ ID NO: 61 and the light chain polypeptide of SEQ ID NO: 56. 
     
     
         21 . A vector comprising the nucleic acid of  claim 1 . 
     
     
         22 . A vector comprising the nucleic acid of  claim 12 . 
     
     
         23 . A vector comprising the nucleic acid of  claim 19 . 
     
     
         24 . A vector comprising the nucleic acid of  claim 20 . 
     
     
         25 . A recombinant cell which comprises a nucleic acid according to  claim 1 , operably linked to a promoter. 
     
     
         26 . A recombinant cell which comprises a nucleic acid according to  claim 12 , operably linked to a promoter. 
     
     
         27 . A recombinant cell which comprises a nucleic acid according to  claim 19 , operably linked to a promoter. 
     
     
         28 . A recombinant cell which comprises a nucleic acid according to  claim 20 , operably linked to a promoter. 
     
     
         29 . A recombinant cell according to  claim 25 , which comprises a mammalian cell. 
     
     
         30 . A recombinant cell according to  claim 28 , which comprises a mammalian cell. 
     
     
         31 . A recombinant cell according to  claim 25 , which comprises a CHO cell. 
     
     
         32 . A recombinant cell according to  claim 28 , which comprises a CHO cell. 
     
     
         33 . A method of producing an antibody or antibody fragment encoded by the nucleic acid of  claim 1  comprising culturing an isolated or recombinant cell comprising said nucleic acid operably linked to a promoter under conditions that result in the expression of said antibody or antibody fragment.

A method of producing an antibody or antibody fragment encoded by the nucleic acid of  claim 12  comprising culturing an isolated or recombinant cell comprising said nucleic acid operably linked to a promoter under conditions that result in the expression of said antibody or antibody fragment.

A method of producing an antibody or antibody fragment encoded by the nucleic acid of  claim 19  comprising culturing an isolated or recombinant cell comprising said nucleic acid operably linked to a promoter under conditions that result in the expression of said antibody or antibody fragment.

A method of producing an antibody or antibody fragment encoded by the nucleic acid of  claim 20  comprising culturing an isolated or recombinant cell comprising said nucleic acid operably linked to a promoter under conditions that result in the expression of said antibody or antibody fragment.
