Abstract

The invention relates to substituted aryl and heteroaryl (R)-Chiral Halogenated 1-Substitutedamino-(n+1)-Alkanol compounds useful as inhibitors of cholesteryl ester transfer protein (CETP; plasma lipid transfer protein-I) and compounds, compositions and methods for treating atherosclerosis and other coronary artery diseases. Novel high yield, stereoselective processes for the preparation of the chiral substituted alkanol compounds from chiral and achiral intermediates are described. Preferred (R)-Chiral 1-Substitutedamino-(n+1)-Alkanol compounds are substituted (R)-Chiral fused heterocyclic amino compounds. A preferred specific (R)-Chiral fused heterocyclic amino compound is:

         68 . A compound of Formula I:  
       
         
           
           
               
               
           
         
       
       or a pharmaceutically acceptable salt thereof, wherein; 
 R 16  is bonded together with a substituent selected from the group consisting of R 4 , R 8 , R 9 , and R 13  to form a heterocyclyl ring having from 6 through 10 members;  
 n is 1 or 2;  
 R 1  is haloalkyl or haloalkoxyalkyl with the proviso that R 1  is selected to have the highest Cahn-Ingold-Prelog stereochemical system ranking of three groups bonded to the hydroxy-substituted carbon to which R 1  and R 2  are attached in radical Ia:  
                     
 which radical Ia is a fragment of Formula I;  
 R 2  is selected from the group consisting of hydrido, aryl, aralkyl, alkyl, alkenyl, alkenyloxyalkyl, haloalkyl, haloalkenyl, halocycloalkyl, haloalkoxyalkyl, haloalkenyloxyalkyl, halocycloalkoxy, halocycloalkoxyalkyl, perhaloaryl, perhaloaralkyl, perhaloaryloxyalkyl, heteroaryl, dicyanoalkyl, and carboalkoxycyanoalkyl;

         70 . Compound of  claim 69  or a pharmaceutically acceptable salt thereof, wherein; 
 R 16  is bonded together with R 4  or R 9  to form a heterocyclyl ring having from 6 through 8 members;

 R 1  is haloalkyl or haloalkoxyalkyl with the proviso that R 1  is selected to have the highest Cahn-Ingold-Prelog stereochemical system ranking of said three groups bonded to the hydroxy-substituted carbon to which R 1  and R 2  are attached in said fragment of the Formula I and with the further proviso that said haloalkyl has two or more halo substituents;  
 R 2  is hydrido;  
 R 3  is hydrido;  
 Y is selected from the group consisting of a bond, CH 2 , and CH 2 CH 2 ;  
 Z is selected from the group consisting of a bond, CH 2 , and CH 2 CH 2 ;  
 R 4  is hydrido or halo unless R 4  is bonded together with R 16 ;  
 R 9  is hydrido or halo unless R 9  is bonded together with R 16 ;
