Closed cell culture system
Abstract
An object of the present invention is to provide a tissue cell culture system whereby a call tissue can be efficiently and quickly proliferated in vivo and the onset bacterial infection in an injured part can be avoided in the course of a treatment. More specifically, a closed cell culture system (1) characterized in that a defection (2) of a tissue on the body surface or inside the body is tightly sealed to form a closed environment free from the invasion of bacteria, etc. and then a solution appropriate for cell culture is circulated in the tissue defection thus sealed to thereby regenerate the defective tissue; and a method of administering a drug which comprises dissolving a remedy in the perfusion with the use of the above system and thus promoting the treatment of the defection.

11 . A closed cell culture system characterized in that a defection of a tissue on the body surface or inside the body is tightly sealed from the outside to form a closed environment free from the invasion of bacteria, and a solution appropriate for cell culture is circulated or perfused in the tissue defection thus sealed to thereby regenerate the defective tissue.
12 . The closed cell culture system according to claim 11 , wherein said defection of a tissue on the body surface is tightly sealed by an adhesive sheet to form said closed environment.
13 . The closed cell culture system according to claim 11 , wherein a bioabsorbable material is embedded in the closed environment to promote regeneration of the defective tissue cells.

The closed cell culture system according to claim 11 , wherein the solution appropriate for cell culture is serum isolated from the patient's blood, a blood preparation, platelet concentrated serum, a serum fraction preparation, a blood protein fraction component solution, a plasma expander, an osmotic pressure isotonic infusion or a cell culture medium.

The closed cell culture system according to claim 11 , wherein a cell proliferation environment suited to analgesia and disinfection of the defection and to each tissue to be regenerated is constructed and programmed by varying the components of the circulating solution at each stage of treatment.

The closed cell culture system according to claim 11 , wherein a gas exchanger connected to a monitoring device which measures pH, carbon dioxide partial pressure and oxygen partial pressure in the closed environment is incorporated into a circuit connected to the closed environment, and the cell culture environment is optimized by automatically adjusting the gas concentrations based on signals from the monitoring unit.
17 . The closed cell culture system according to claim 11 , wherein the pressure inside the closed environment is continuously or intermittently controlled.
18 . The closed cell culture system according to claim 11 , comprising a circuit for perfusing a perfusion the osmotic pressure of which has been adjusted externally.

A combination of a perfusion introduction tube, a perfusion discharge tube, and a disc used for the system according to claim 11 , wherein said perfusion introduction tube introduces said cell culture solution in said defection of a tissue on the body surface, said perfusion discharge tube discharges the introduced fluid, and said disc comprises disc 1 and disc 2 , said perfusion introduction tube being provided in the disc 2 , perfusion from said perfusion introduction tube being introduced into said defection of a tissue via said disc 2 , and said perfusion discharge tube being provided in said disc 1 , whereby perfusion filling said defection of a tissue passes through an airspace between said disc 1 and disc 2 and is discharged from said perfusion discharge tube, thereby completing perfusion of cell culture solution into said defection of a tissue.
20 . The combination according to claim 19 , further comprising a bioabsorbable material for epidermis, wherein said perfusion is introduced into said defection by absorption into the material via said disc 2 .

The combination according to claim 20 , further comprising an embedded bioabsorbable material which is to be embedded in said defection, wherein said perfusion absorbed into the bioabsorbable material for epidermis passes through said airspace after further absorption into the embedded bioabsorbable material.
22 . The closed cell culture system according to claim 11 , wherein a cell culture space is created within an organ or deep inside the body cavity through a combination of a puncture device and a balloon catheter.
23 . A method of administering a drug which promotes treatment of a defection wherein a therapeutic drug is dissolved in a perfusion using the closed cell culture system according to claim 11 .
24 . A method of cell transport and proliferation for promoting healing of the injured part, whereby cells from the patient are first collected, cultured and proliferated in vitro, then suspended in a perfusion and returned to the closed culture system via the perfusion system so as to provide therapy by means of a cell culture of the injured part while, in conjunction therewith, culture proliferation and take are performed by adding tissue cultured in vitro.Join the waitlist — get patent alerts
