Cell migration assay

Abstract

The present invention is directed to a modified cell migration assay allowing for improved identification and discrimination of chemokine receptor antagonists from non-specific blockers.

         21 . A kit, comprising: 
 a cell migration apparatus; and    a solution comprising an inhibitory concentration of ligand for a chemoattractant receptor-bearing cell.    
     
     
         22 . The kit of  claim 21 , wherein the solution is lyophilized.  
     
     
         23 . The kit of  claim 21 , wherein the chemoattractant receptor comprises FPRL-1 receptor and the ligand is selected from the group consisting of w-peptide 1 and w-peptide 2.  
     
     
         24 . The kit of  claim 21 , wherein the chemoattractant receptor comprises C5A receptor, and the ligand comprises C5A.  
     
     
         25 . The kit of  claim 21 , further comprising the chemoattractant receptor-bearing cell.  
     
     
         26 . The kit of  claim 21 , further comprising instructional materials.  
     
     
         27 . The kit of  claim 21 , wherein the inhibitory concentration of a ligand for the chemoattractant receptor inhibits cell migration greater than or equal to 50% of maximal ligand-activated cell migration.  
     
     
         28 . The kit of  claim 21 , wherein the inhibitory concentration of a ligand for the chemoattractant receptor inhibits cell migration greater than or equal to 85% of maximal ligand-activated cell migration.  
     
     
         29 . The kit of  claim 21 , wherein the inhibitory concentration of a ligand for the chemoattractant receptor inhibits cell migration greater than or equal to 95% of maximal ligand-activated cell migration.  
     
     
         30 . The kit of  claim 21 , wherein the chemoattractant receptor is a chemokine receptor and the ligand for the chemoattractant receptor is a chemokine.  
     
     
         31 . The kit of  claim 30 , wherein the chemokine receptor is CXCR1, CXCR2, CXCR3, CXCR4, CXCR5, CXCR6, CCR1, CCR2, CCR3, CCR4, CCR5, CCR6, CCR7, CCR8, CCR9, CCR10, CCR11, CX3CR1, XCR1, or homologs thereof.  
     
     
         32 . The kit of  claim 30 , wherein the chemokine is IL-8, GCP-2, Gro α, Groβ, Gro γ, ENA-78, PBP, MIG, IP-10, I-TAC, SDF-1, BLC, MIP-1α, MIP-1β, CXCL16, RANTES, HCC-1, HCC-2, HCC-3, HCC-4, MCP-1, MCP-2, MCP-3, MCP-4, eotaxin-1, eotaxin-2, TARC, MDC, MIP-3α, MIP-3β, 6Ckine, I-309, TECK, lymphotactin, fractalkine, TCA-4, Exodus-2, Exodus-3, CKβ-11, active variants, or homologs thereof.  
     
     
         33 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR5, and the chemokine comprises MIP-1α, MIP-1β, or RANTES.  
     
     
         34 . The kit of  claim 30 , wherein the chemokine receptor comprises CXCR4, and the chemokine comprises SDF-1.  
     
     
         35 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR7, and the chemokine comprises MIP3β or 6Ckine.  
     
     
         36 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR9, and the chemokine comprises TECK.  
     
     
         37 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR6, and the chemokine comprises MIP-3α.  
     
     
         38 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR10, and the chemokine comprises CTACK or MEC.  
     
     
         39 . The kit of  claim 30 , wherein the chemokine receptor comprises CCR2, and the chemokine comprises MCP-1, MCP-2 or MCP-4.  
     
     
         40 . The kit of  claim 30 , wherein the chemokine receptor comprises CXCR3, and the chemokine comprises I-TA, IP-10. or MIG.

 a cell migration apparatus;    a chemoattractant receptor-bearing cell;    a solution comprising an inhibitory concentration of ligand for a chemoattractant receptor-bearing cell; and    instructional materials.
