Claims

Abstract

The invention relates to a composition comprising at least one collagenolytic active enzyme with enzymatic activity at acidic pH for drilless enzymatic caries removal. Furthermore, the present invention relates to a process of producing this collagenolytic active enzyme comprising composition and to processes of removing caries. The invention also relates to the use of collagenolytic active enzyme comprising compositions for producing a treatment agent for dental applications.

Composition for the treatment of dental caries comprising at least one collagenolytic active enzyme, comprising serine-carboxyl proteinase, at least one solvent and at least one acid, wherein said composition has a collagenolytic activity of more than 5 U/ml and a pH value of about 1 to about 5.  
   
   
       2 . Composition according to  claim 1 , wherein said at least one collagenolytic active enzyme is selected from the group consisting of Kumamolysin and ScP-A.  
   
   
       3 . Composition according to  claim 1 , wherein said at least one collagenolytic active enzyme is obtainable from  Bacillus novosp.  MN-32 or  Alicyclobacillus sendaiensis  strain NTAP- 1.  
   
   
       4 . Composition according to  claim 1 , wherein said composition comprises at least one further proteinase.  
   
   
       5 . Composition according to  claim 4 , wherein one of said further proteinases is pepsin.  
   
   
       6 . Composition according to  claim 1 , wherein said composition comprises at least one additive.  
   
   
       7 . Composition according to  claim 2 , wherein said composition comprises at least one additive.  
   
   
       8 . Composition according to  claim 4 , wherein said composition comprises at least one additive.  
   
   
       9 . Composition according to  claim 1 , wherein said at least one collagenolytic active enzyme is thermally stable.  
   
   
       10 . Composition according to  claim 1 , wherein at least one collagenolytic active enzyme is activated by Ca-ions.

Process of preparation a composition for the treatment of dental caries according to  claim 1 , wherein said at least one collagenolytic active enzyme, a solvent and at least one acid are mixed so that said composition has an collagenolytic activity of more than 5 U/ml and a pH value of about 1 to about 5.

Kit comprising at least one collagenolytic active enzyme comprising serine-carboxyl proteinase, at least one solvent and at least one acid, wherein said at least one collagenolytic active enzyme, said at least one solvent and said at least one acid are miscible for obtaining a composition according to  claim 1 .  
   
   
       13 . Composition comprising at least one collagenolytic active enzyme comprising serine-carboxyl proteinase for dental applications.  
   
   
       14 . Composition according to  claim 13 , wherein said at least one collagenolytic active enzyme is of non-animal origin.  
   
   
       15 . Composition according to  claim 13 , wherein said at least one collagenolytic active enzyme is selected from the group consisting of Kumamolysin and ScP-A.  
   
   
       16 . Composition according to  claim 13 , wherein said at least one collagenolytic active enzyme is obtainable from  Bacillus novosp.  MN-32 or  Alicyclobacillus sendaiensis  strain NTAP- 1.  
   
   
       17 . Composition according to  claim 13 , wherein said composition comprises at least one further proteinase.

       20 . Composition according to  claim 20 , wherein said at least one with additive is selected from the group consisting of rheological additives, inhibitors and buffers.

Kit comprising at least one collagenolytic active enzyme comprising serine-carboxyl proteinase, at least one solvent and at least one acid, wherein said at least one collagenolytic active enzyme, said at least one solvent and said at least one acid are miscible for obtaining a composition according to  claim 13 .  
   
   
       22 . Method of caries removal comprising the step of applying a composition according to  claim 1 .  
   
   
       23 . Method of caries removal further comprising the step of penetrating tooth enamel by treating a portion of the tooth with a mechanical tool before applying a composition according to  claim 1  to the treated tooth portion.  
   
   
       24 . Method according to  claim 22 , further comprising the step of removing an existing filling before applying the composition according to  claim 1  to the tooth.  
   
   
       25 . Method according to  claim 22 , further comprising the step of treating the tooth area affected by caries with a rotating drill before applying the composition of  claim 1  to the affected tooth area.  
   
   
       26 . Multi chamber device for storing and dispensing compositions according to  claim 1 , wherein at least one chamber contains a collagenolytic active enzyme comprising serine-carboxyl proteinase.  
   
   
       27 . Multi chamber device according to  claim 26 , wherein said at least one chamber that does not contain a collagenolytic active enzyme comprising serine-carboxyl proteinase comprises a substance providing for a pH value at which the collagenolytic active enzyme, after mixing the contents of the chambers, is most active.
