Abstract

The invention provides polypeptides that act both as an agonist of the GLP-1 receptor and an antagonist of the glucagon receptor. Such polypeptides are useful for treating individuals with type 2 diabetes or other metabolic disorders.

A method of treating or preventing a disease or condition selected from the group consisting of diabetes (Type 2), maturity-onset diabetes of the young (MODY), latent autoimmune diabetes adult (LADA), impaired glucose tolerance (IGT), impaired fasting glucose (IFG), gestational diabetes, and metabolic syndrome X, comprising administering to a mammal an effective amount of a peptide that is a GLP-1 receptor agonist and a glucagon antagonist.  
     
     
         5 . The method of  claim 4 , wherein the peptide is selected from the group consisting of SEQ ID NOs: 6 through 32, and functionally equivalent fragments, derivatives and variants thereof.  
     
     
         6 . The method of  claim 5 , wherein the peptide is selected from the group consisting of SEQ ID NO: 25, SEQ ID NO: 26, and SEQ ID NO: 27.  
     
     
         7 . The method of  claim 4 , further comprising administering an α-glucosidase inhibitor with said peptide.
