Abstract

The present invention relates to polynucleotide and polypeptide molecules for zdint1, a novel member of the Disintegrin Proteases. The polypeptides, and polynucleotides encoding them, are believed to be cell-cell interaction modulating and may be used for delivery and therapeutics. The present invention also includes antibodies to the zdint1 polypeptides.

An isolated polypeptide molecule comprising a contiguous sequence of 13 amino acids of SEQ ID NO:2, wherein the contiguous sequence contains an integrin binding component having a contiguous amino acid sequence of Glu Cys Asp as shown in SEQ ID NO:2 from residue 443 to residue 445, and wherein the polypeptide binds an integrin.  
     
     
         2 . The isolated polypeptide molecule according to  claim 1 , wherein the polypeptide molecule comprises a contiguous sequence of 14 amino acids of SEQ ID NO:2.  
     
     
         3 . The isolated polypeptide molecule according to  claim 1 , wherein the polypeptide molecule comprises residues 437 to 450 of SEQ ID NO:2.  
     
     
         4 . The isolated polypeptide molecule according to  claim 1 , wherein the polypeptide molecule is between 82 and 232 amino acids in length.  
     
     
         5 . An isolated polypeptide molecule comprising a disintegrin loop having a contiguous amino acid sequence of residues 438 to 449 as shown in SEQ ID NO:2, and wherein the polypeptide molecule binds an integrin.  
     
     
         6 . The polypeptide molecule according to  claim 5 , wherein the disintegrin loop consists of residues 438 to 449 as shown in SEQ ID NO:2.  
     
     
         7 . An isolated polypeptide, consisting of residues 438 to 449 as shown in SEQ ID NO:2.  
     
     
         8 . An isolated polypeptide, consisting of residues 437 to 450 as shown in SEQ ID NO:2.  
     
     
         9 . An isolated polynucleotide molecule encoding a polypeptide molecule, wherein the polypeptide molecule comprises a contiguous sequence of 14 amino acids of SEQ ID NO:2.  
     
     
         10 . An isolated polynucleotide molecule according to  claim 9 , wherein the polypeptide molecule comprises residues 437 to 450 of SEQ ID NO:2.  
     
     
         11 . A method for modulating cell-cell interactions by combining the polypeptide according to  claim 1 , with cells in vivo and in vitro.  
     
     
         12 . The method for modulating cell-cell interactions according to  claim 11 , whereby the cells are derived from tissues selected from the group consisting of: 
 a) tissues from heart;    b) tissues from brain;    c) tissues from spinal cord; and    d) tissues from skeletal muscle.    
     
     
         13 . An isolated polypeptide molecule comprising residues 465 to 696 of SEQ ID NO:2.  
     
     
         14 . An antibody that specifically binds to a polypeptide comprising the amino acid sequence of SEQ ID NO:2 or to a fragment of said polypeptide wherein the fragment consists of an amino acid sequence selected from the group consisting of: 
 (a) residue 1 to residue 696 of SEQ ID NO:2;    (b) residue 1 to residue 163 of SEQ ID NO:2;    (c) residue 164 to residue 382 of SEQ ID NO:2;    (d) residue 383 to residue 464 of SEQ ID NO:2;    residue 465 to residue 696 of SEQ ID NO:2;    residue 438 to residue 449 of SEQ ID NO:2, and    residue 437 to 450 of SEQ ID NO:2.    
     
     
         15 . The antibody according to  claim 14 , wherein the antibody is a monoclonal antibody.  
     
     
         16 . A method of producing an antibody comprising the following steps in order: 
 inoculating an animal with a polypeptide selected from the group consisting of:    (a) residue 1 to residue 696 of SEQ ID NO:2;    (b) residue 1 to residue 163 of SEQ ID NO:2;    (c) residue 164 to residue 382 of SEQ ID NO:2;    (d) residue 383 to residue 464 of SEQ ID NO:2;    residue 465 to residue 696 of SEQ ID NO:2;    residue 438 to residue 449 of SEQ ID NO:2, and    residue 437 to 450 of SEQ ID NO:2    wherein the polypeptide elicits an immune response in the animal to produce the antibody, and isolating the antibody from the animal;    and wherein the antibody specifically binds to the amino acid sequence of SEQ ID NO:2 from residue 1 to residue 696.    
     
     
         17 . An antibody produced by the method of  claim 16  which specifically binds to residues 1 to 696 of SEQ ID NO:2.  
     
     
         18 . The antibody produced by the method of  claim 16 , wherein the antibody is a monoclonal antibody.  
     
     
         19 . An antibody which specifically binds to a polypeptide of amino acid residues 1 to 696 of SEQ ID NO:2.
