Abstract

The enzyme Lp-PLA 2 in purified form, an isolated nucleic acid molecule encoding Lp-PLA 2 , the use of an inhibitor of the enzyme Lp-PLA2 in therapy and a method of screening compounds to identify those compounds which inhibit the enzyme.

         25 . Isolated Lp-PLA2 made by a method for isolating Lp-PLA2 comprising the steps of: 
 growing cells comprising an Lp-PLA2 gene under conditions whereby Lp-PLA2 protein is expressed, and    isolating Lp-PLA2 protein from the cells or growth media.    
     
     
         26 . The method of  claim 25  wherein the cells are host cells.  
     
     
         27 . The method of  claim 26  wherein the host comprises a vector comprising a sequence encoding Lp-PLA2.  
     
     
         28 . The method of  claim 25  wherein the Lp-PLA2 is at least 80% pure.  
     
     
         29 . The method of  claim 25  wherein the Lp-PLA2 is at least 90% pure.  
     
     
         30 . The method of  claim 25  wherein the Lp-PLA2 is at least 95% pure.  
     
     
         31 . The method of  claim 25  wherein the Lp-PLA2 is at least 99% pure.  
     
     
         32 . The method of  claim 25  wherein the Lp-PLA2 gene encodes an amino acid sequence comprising at least one amino selected from the group of SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID No: 10 and SEQ ID NO: 11.  
     
     
         33 . The method of  claim 25  wherein the Lp-PLA2 is expressed from a nucleic acid sequence comprising at least one sequence selected from the group of SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7, and SEQ ID NO: 8 and SEQ ID NO: 9  
     
     
         34 . Isolated Lp-PLA2 made by a method for isolating Lp-PLA2 comprising the steps of: 
 obtaining a human sample comprising Lp-PLA2 protein, and isolating Lp-PLA2 protein from the sample.    
     
     
         35 . The method of  claim 34  wherein the sample comprises cells.  
     
     
         36 . The method of  claim 34  wherein the sample comprises a bodily fluid.  
     
     
         37 . The method of  claim 34  wherein the Lp-PLA2 is at least 80% pure.  
     
     
         38 . The method of  claim 34  wherein the Lp-PLA2 is at least 90% pure.  
     
     
         39 . The method of  claim 34  wherein the Lp-PLA2 is at least 95% pure.  
     
     
         40 . The method of  claim 34  wherein the Lp-PLA2 is at least 99% pure.  
     
     
         41 . The method of  claim 34  wherein the sample comprises at least one of a composition of matter selected from the group of blood, a subfraction of blood, a subfraction of LDL, and plasma.  
     
     
         42 . The method of  claim 41  wherein the sample comprises blood.  
     
     
         43 . The method of  claim 41  wherein the sample comprises a subfraction of blood.  
     
     
         44 . The method of  claim 41  wherein the sample comprises a subfraction of LDL.
