Abstract

Human anti-epidermal growth factor receptor (EGFR) single-chain antibodies (scFvs) were isolated from a human IgM phage display library using purified epidermal growth factor receptor as antigen. Two isolates with different amino acid sequences were identified by ELISA as epidermal growth factor receptor-specific. The scFvs bind to the full length epidermal growth factor receptor and the truncated and/or mutated epidermal growth factor receptor on human cells. These anti-EGFR-scFvs are useful as therapeutic and/or diagnostic agents.

Claims

exact text as granted — not AI-modified
1 . An isolated human anti-epidermal growth factor receptor single-chain antibody having the sequence of SEQ ID No. 1 or biologically functional fragment thereof, wherein said antibody binds to the extracellular domain of epidermal growth factor receptor.

2 . A pharmaceutical composition comprising the anti-epidermal growth factor receptor single-chain antibody of claim 1 and a pharmaceutically acceptable carrier.

3 . The pharmaceutical composition of claim 2 , further comprising a therapeutic agent.

4 . The pharmaceutical composition of claim 3 , wherein said therapeutic agent is a toxin, a chemotherapeutic agent, a radioisotope, or a gene therapy vector.

5 . The pharmaceutical composition of claim 2 , further comprising a diagnostic agent.

6 . The pharmaceutical composition of claim 5 , wherein said diagnostic agent is a radioisotope or a transition metal.

         7 . A method of treating a tumor, comprising:
administering to a patient in need of such treatment an effective amount of an anti-epidermal growth factor receptor single-chain antibody of claim 1 .

         8 . The anti-epidermal growth factor receptor single-chain antibody of claim 1 , further comprising a radiolabel.

9 . A method for the diagnostic location and assessment of tumor growth, comprising:
administering to a patient in need of such diagnostic treatment an effective amount of an anti-epidermal growth factor receptor single-chain antibody of claim 8 .

10 . An isolated DNA encoding the anti-epidermal growth factor receptor single-chain antibody of claim 1 .

11 . An expression vector comprising the isolated DNA of claim 10 .

12 . A human anti-epidermal growth factor receptor single-chain antibody having the sequence of SEQ ID No. 2 or biologically functional fragment thereof, wherein said antibody binds to the extracellular domain of epidermal growth factor receptor.

13 . A pharmaceutical composition comprising the anti-epidermal growth factor receptor single-chain antibody of claim 12 and a pharmaceutically acceptable carrier.

14 . The pharmaceutical composition of claim 13 , further comprising a therapeutic agent.

15 . The pharmaceutical composition of claim 14 , wherein said therapeutic agent is a toxin, a chemotherapeutic agent, a radioisotope, or a gene therapy vector.

16 . The pharmaceutical composition of claim 13 , further comprising a diagnostic agent.

17 . The pharmaceutical composition of claim 16 , wherein said diagnostic agent is a radioisotope or a transition metal.

         18 . A method of treating a tumor, comprising:
administering to a patient in need of such treatment an effective amount of an anti-epidermal growth factor receptor single-chain antibody of claim 12 .

         19 . The anti-epidermal growth factor receptor single-chain antibody of claim 12 , further comprising a radiolabel.

20 . A method for the diagnostic location and assessment of tumor growth, comprising:
administering to a patient in need of such diagnostic treatment an effective amount of an anti-epidermal growth factor receptor single-chain antibody of claim 19 .

21 . An isolated DNA encoding the anti-epidermal growth factor receptor single-chain antibody of claim 12 .

22 . An expression vector comprising the isolated DNA of claim 21.
