Abstract

A method of immunotherapy to treat cancer or a synergistic anti-cancer treatment by administering an effective amount of a natural cytokine mixture (NCM), an effective amount of cyclophosphamide (CY), or an effective amount of indomethacin (INDO), wherein the NCM, CY, or INDO are administered singly or in combinations thereof. An anti-metastatic treatment method by promoting differentiation and maturation of immature dendritic cells in a lymph node; allowing presentation thereof; and preventing development of metastasis. A method of using an NCM as a diagnostic skin test for predicting treatment outcome. A method of pre-treating dendritic cells (DC) and a method of treating monocyte defects characterized by sinus histiocytosis or a negative NCM skin test. Compositions and methods for eliciting an immune response to endogenous or exogenous tumor antigens.

         33 . An anti-cancer composition comprising an effective amount of a chemical inhibitor and an effective amount of a non-steroidal anti-inflammatory drug (NSAID).  
     
     
         34 . The composition according to  claim 33 , wherein said chemical inhibitor is an anti-neoplastic agent.  
     
     
         35 . The composition according to  claim 33 , wherein said chemical inhibitor is cyclophosphamide (CY).  
     
     
         36 . The composition according to  claim 33 , wherein said NSAID is selected from the group consisting of indomethacin (INDO), Ibuprofen, celecoxib (Celebrex®), rofecoxib (Vioxx®), CoxII inhibitors, and combinations thereof.  
     
     
         37 . The composition according to  claim 33 , wherein said NSAID is INDO.  
     
     
         38 . A method of immunotherapy to treat cancer comprising the step of administering an effective amount of a chemical inhibitor and an effective amount of an NSAID.  
     
     
         39 . The method according to  claim 38 , wherein said chemical inhibitor is an anti-neoplastic agent.  
     
     
         40 . The method according to  claim 38 , wherein said chemical inhibitor is cyclophosphamide (CY).  
     
     
         41 . The method according to  claim 38 , wherein said NSAID is selected from the group consisting of indomethacin (INDO), Ibuprofen, celecoxib (Celebrex®), rofecoxib (Vioxx®), CoxII inhibitors, and combinations thereof.  
     
     
         42 . The method according to  claim 38 , wherein said NSAID is INDO.  
     
     
         43 . The method according to  claim 38 , wherein the chemical inhibitor is administered prior to the NSAID.  
     
     
         44 . The method according to  claim 38 , wherein the chemical inhibitor is administered at a dose of about 300 mg/m 2 .  
     
     
         45 . The method according to  claim 38 , wherein zinc is additionally administered.  
     
     
         46 . The method according to  claim 38 , wherein the NSAID is administered at a dose of about 25 mg, orally, three times daily.  
     
     
         47 . An anti-cancer composition comprising an effective amount of a chemical inhibitor in combination with an effective amount of an NSAID and an effective amount of IFN-γ, IL-2, IL-1, and TNF-α, wherein said IFN-γ, IL-2, IL-1, and TNF-α can be naturally made, recombinants thereof, or a mixture of naturally made and recombinants thereof.  
     
     
         48 . The composition according to  claim 47 , wherein said chemical inhibitor is an anti-neoplastic agent.  
     
     
         49 . The composition according to  claim 47 , wherein said chemical inhibitor is cyclophosphamide (CY).  
     
     
         50 . The composition according to  claim 47 , wherein said NSAID is selected from the group consisting of indomethacin (INDO), Ibuprofen, celecoxib (Celebrex®), rofecoxib (Vioxx®), CoxII inhibitors, and combinations thereof.  
     
     
         51 . The composition according to  claim 47 , wherein said NSAID is INDO.  
     
     
         52 . A method of immunotherapy to treat cancer comprising the step of administering an effective amount of a chemical inhibitor in combination with an effective amount of an NSAID and an effective amount of IFN-γ, IL-2, IL-1, and TNF-α, wherein said IFN-γ, IL-2, IL-1, and TNF-α can be naturally made, recombinants thereof, or a mixture of naturally made and recombinants thereof.  
     
     
         53 . The method according to  claim 52 , wherein said chemical inhibitor is an anti-neoplastic agent.  
     
     
         54 . The method according to  claim 52 , wherein said chemical inhibitor is cyclophosphamide (CY).  
     
     
         55 . The method according to  claim 52 , wherein said NSAID is selected from the group consisting of indomethacin (INDO), Ibuprofen, celecoxib (Celebrex®), rofecoxib (Vioxx®), CoxII inhibitors, and combinations thereof.  
     
     
         56 . The method according to  claim 52 , wherein said NSAID is INDO.  
     
     
         57 . An anti-cancer treatment method comprising the steps of administering an effective amount of a chemical inhibitor and an NSAID in combination with an effective amount of a cytokine mixture including IL-1, IL-2, IL-6, IL-8, IFN-γ, and TNF-α, wherein said cytokines can be naturally made, recombinants thereof, or a mixture of naturally made and recombinants thereof.  
     
     
         58 . The method according to  claim 57 , wherein said chemical inhibitor is an anti-neoplastic agent.  
     
     
         59 . The method according to  claim 57 , wherein said chemical inhibitor is cyclophosphamide (CY).  
     
     
         60 . The method according to  claim 57 , wherein said NSAID is selected from the group consisting of indomethacin (INDO), Ibuprofen, celecoxib (Celebrex®), rofecoxib (Vioxx®), CoxII inhibitors, and combinations thereof.

         62 . The method according to  claim 57 , wherein said cytokine mixture further comprises IL-12, GM-CSF or G-CSF or combinations thereof.
