Abstract

DNA vaccine against a pathogen affecting farm animals, in particular bovines or porcines, comprising a plasmid containing a nucleotide sequence encoding an immunogen of a pathogen of the animal species considered, under conditions allowing the in vivo expression of this sequence, and a cationic lipid containing a quaternary ammonium salt, of formula in which R1 is a saturated or unsaturated linear aliphatic radical having 12 to 18 carbon atoms, R2 is another aliphatic radical containing 2 or 3 carbon atoms, and X a hydroxyl or amine group, this lipid being preferably DMRIE.

A DNA vaccine against a porcine pathogen, wherein the porcine pathogen is porcine herpesvirus or Aujeszky's disease (pseudorabies virus or PRV), porcine reproductive respiratory syndrome virus (or PRRSV), swine influenza virus (or SIV), conventional hog cholera virus (or HCV) or a parvovirus in porcines, comprising at least one plasmid that contains and expresses in a porcine host cell a nucleotide sequence encoding an immunogen of the porcine pathogen, and a cationic lipid containing a quaternary ammonium salt, of the formula

         113 . The vaccine according to  claim 110 , wherein the immunogen is PRV gD, modified by substitution of the PRV gD signal sequence with a human tPA signal sequence, and/or by deletion of the transmembrane domain.

The vaccine according to  claim 85  comprising a first plasmid that contains and expresses in a porcine host cell a nucleotide sequence encoding porcine pseudorabies virus glycoprotein gB (PRV gB), modified by substitution of the PRV gB signal sequence with a human tPA signal sequence and deletion of the transmembrane domain and contiguous C-terminal portion; a second plasmid that contains and expresses in a porcine host cell a nucleotide sequence encoding PRV gC, modifiedby substitution of the PRV gC signal sequence with a human tPA signal sequence and deletion of the transmembrane domain and contiguous C-terminal portion and a third plasmid that contains and expresses in a porcine host cell a nucleotide sequence encoding PRV gD, modified by substitution of the PRV gD signal sequence with a human tPA signal sequence and deletion of the transmembrane domain and contiguous C-terminal portion; and wherein the lipid is DMRIE, whereby the vaccine comprises DMRIE-DOPE.
