Abstract

Pharmaceutical compositions comprising tricyclic 3,4-propinoperhydropurines and uses thereof for blocking neuronal transmission are provided.

A method of interfering with neuronal transmission comprising contacting a neuron with an effective amount of the composition comprising at least one compound selected from the group consisting of Gonyautoxin 1, Gonyautoxin 2, Gonyautoxin 3, Gonyautoxin 4 and Gonyautoxin 5; and a pharmaceutically acceptable carrier.  
   
   
       2 . The method of  claim 1 , wherein the composition comprises a mixture of at least two compounds selected from the group consisting of Gonyautoxin 1, Gonyautoxin 2, Gonyautoxin 3, Gonyautoxin 4 and Gonyautoxin 5.  
   
   
       3 . The method of  claim 1 , wherein the composition further comprises Botulin A toxin.  
   
   
       4 . The method of  claim 1 , wherein about 100 to about 800 microliters of said composition is contacted with said neuron.  
   
   
       5 . The method of  claim 1 , wherein less than one milliliter of said composition is contacted with said neuron.  
   
   
       6 . The method of  claim 1 , wherein said contacting step comprises injecting the composition into an area proximate to the neuron.  
   
   
       7 . The method of  claim 1 , wherein said composition comprises about 20 to about 40 units of the compound per milliliter of total composition.  
   
   
       8 . The method of  claim 1 , wherein the carrier is acetic acid.  
   
   
       9 . The method of  claim 1 , wherein said neuron is innervating a muscle.  
   
   
       10 . The method of  claim 9 , wherein said muscle is inside a human face.  
   
   
       11 . A method as claimed in  claim 1 , wherein the composition consists of Gonyautoxin 2, Gonyautoxin 3 and a pharmaceutically acceptable carrier.  
   
   
       12 . A method interfering with muscle contraction comprising contacting a muscle with an effective amount of a composition comprising at least one compound selected from the group consisting of Gonyautoxin 1, Gonyautoxin 2, Gonyautoxin 3, Gonyautoxin 4, and Gonyautoxin 5; and a pharmaceutically acceptable carrier.  
   
   
       13 . The method of  claim 12 , wherein the composition comprises a mixture of Gonyautoxin 2 and Gonyautoxin 3.  
   
   
       14 . The method of  claim 12 , wherein the composition further comprises Botulin A toxin.  
   
   
       15 . The method of  claim 12 , wherein the composition comprises about 20 to about 40 units of the compound per milliliter of the carrier.  
   
   
       16 . The method of  claim 12 , wherein the carrier is acetic acid.  
   
   
       17 . The method of  claim 12 , wherein the composition comprises about twenty to about forty units of the compound per milliliter of total composition.  
   
   
       18 . The method of  claim 12 , wherein contacting an effective amount of the composition with a muscle comprises injecting the composition into an area proximate to the muscle.  
   
   
       19 . The method of  claim 18 , wherein no more than one milliliter: of the composition is injected into an area proximate to the muscle per injection point.  
   
   
       20 . The method of  claim 19 , wherein about 100 to about 800 microliters of the composition is injected into an area proximate to the muscle per injection point.  
   
   
       21 . The method of  claim 12 , wherein the muscle is inside a human face.  
   
   
       22 . The method of  claim 12 , wherein the composition consists of Gonyautoxin 2, Gonyautoxin 3 and a pharmacologically acceptable carrier.  
   
   
       23 . A pharmaceutical composition for interfering with neuronal transmission, the composition consisting of an effective amount of Gonyautoxin 2, Gonyautoxin 3 and a pharmacologically acceptable carrier.  
   
   
       24 . The composition of  claim 23 , wherein the composition includes about twenty to about forty units of the Gonyautoxin 2 and Gonyautoxin 3 per milliliter of total solution.  
   
   
       25 . The composition of  claim 23 , wherein the pharmaceutically acceptable carrier is acetic acid.  
   
   
       26 . A preparation for facial rejuvenation comprising an effective amount of a composition comprising at least one compound selected from the group consisting of Gonyautoxin 1, Gonyautoxin 2, Gonyautoxin 3, Gonyautoxin 4, and Gonyautoxin 5; and a facial cream.  
   
   
       27 . The preparation of  claim 26 , wherein said composition comprises about twenty to about forty units of the at least one compound per milliliter of total solution.  
   
   
       28 . The preparation of  claim 26 , further comprising a pharmaceutically acceptable carrier.  
   
   
       29 . The preparation of  claim 28 , wherein said pharmaceutically acceptable carrier is acetic acid.  
   
   
       30 . The preparation of  claim 26  wherein the composition consists of Gonyautoxin 2, Gonyautoxin 3 and a facial cream.  
   
   
       31 . The preparation of  claim 26 , further comprising Botulin A toxin.
