Abstract

The present invention generally relates to nicotinic compounds, in the form of aryl substituted olefinic compounds, as well as pro-drug, N-oxide, metabolite and pharmaceutically acceptable salt forms thereof. Methods of modulating neurotransmitter release via administration of the compounds, pro-drugs, N-oxides and/or pharmaceutically acceptable salts are also disclosed.

         2 . A method of treating a central nervous system disorder comprising administering to a subject in need thereof, an effective amount of a compound denoted (2S)-(4E)-N-methyl-5-(5-methoxy-3-pyridyl)-4-penten-2-amine, or pharmaceutically acceptable salt thereof.  
     
     
         3 . The method of  claim 2 , wherein the disorder is characterized by an alteration in normal neurotransmitter release.  
     
     
         4 . The method of  claim 2 , wherein the disorder is selected from the group consisting of presenile dementia, senile dementia, Lewy Body dementia, Parkinsonism, Huntington's chorea, tardive dyskinesia, hyperkinesia, mania, attention deficit disorder, anxiety, dyslexia, schizophrenia, mild cognitive impairment and Tourette's syndrome.  
     
     
         5 . A method of treating a central nervous system disorder comprising administering to a subject in need thereof, an effective amount of a compound denoted (2S)-(4E)-N-methyl-5-(5-methoxy-3-pyridyl)-4-penten-2-amine, or pharmaceutically acceptable salt thereof.  
     
     
         6 . The method of  claim 5 , wherein the disorder is characterized by an alteration in normal neurotransmitter release.

The method of  claim 5 , wherein the disorder is selected from the group consisting of presenile dementia, senile dementia, Lewy Body dementia, Parkinsonism, Huntington's chorea, tardive dyskinesia, hyperkinesia, mania, attention deficit disorder, anxiety, dyslexia, schizophrenia, mild cognitive impairment and Tourette's syndrome.

A method of treating a central nervous system disorder characterized by an alteration in normal neurotransmitter release comprising administering to a subject in need thereof, an effective amount of a compound denoted (2S)-(4E)-N-methyl-5-(5-methoxy-3-pyridyl)-4-penten-2-amine, or pharmaceutically acceptable salt thereof.  
     
     
         9 . The method of  claim 8 , wherein the disorder is characterized by an alteration in normal neurotransmitter release.

The method of  claim 8 , wherein the disorder is selected from the group consisting of presenile dementia, senile dementia, Lewy Body dementia, Parkinsonism, Huntington's chorea, tardive dyskinesia, hyperkinesia, mania, attention deficit disorder, anxiety, dyslexia, schizophrenia, mild cognitive impairment and Tourette's syndrome.
