Abstract

The invention generally relates to compositions and methods for identifying and testing tyrosine kinase signaling pathway agonists and antagonists, and more particularly, methods and compositions for screening compounds and identifying compounds that will modulate the interaction of protein tyrosine kinase substrates with their intracellular ligands, as well as between their intracellular ligands and other members of the signaling pathway.

A purified antibody which is generated against a polypeptide comprising at least about five non-phosphorylated amino acids of the amino acid sequence of SEQ ID NO: 4 and which specifically binds to a LAT polypeptide comprising an amino acid sequence according to SEQ ID NO: 4.  
     
     
         27 . The purified antibody of  claim 26 , wherein the antibody is polyclonal.  
     
     
         28 . The purified antibody of  claim 27 , wherein the polypeptide comprising at least about five non-phosphorylated amino acids comprises SEQ ID NO: 4.  
     
     
         29 . The purified antibody of  claim 27 , wherein the polypeptide comprises at least about 20 amino acids of SEQ ID NO: 4.  
     
     
         30 . The purified antibody of  claim 29 , wherein said polypeptide comprising at least about twenty amino acids of the non-phosphorylated amino acid sequence of SEQ ID NO: 4 comprises amino acids 31-233 of LAT.  
     
     
         31 . The purified antibody of  claim 27 , wherein the polypeptide comprises SEQ ID NO: 4.  
     
     
         32 . The purified antibody of  claim 27 , wherein said polypeptide comprises the cytostolic tail of LAT.  
     
     
         33 . The purified antibody of any of  claim 27 , wherein the antibody immunoprecipitates a 40 kd protein from a lysate of C305 cells transfected with a LAT cDNA comprising a nucleic acid sequence according to SEQ ID NO: 1, but which does not immunoprecipitate said 40 kd protein from a lysate of non-transfected cells.  
     
     
         34 . The purified antibody of  claim 27 , wherein the antibody does not stain endothelial cells.  
     
     
         35 . The purified antibody of  claim 26 , wherein the antibody is a monoclonal antibody.  
     
     
         36 . The purified antibody of  claim 35 , wherein the polypeptide comprising at least about five non-phosphorylated amino acids comprises SEQ ID NO: 4.  
     
     
         37 . The purified antibody of  claim 35 , wherein the polypeptide comprises at least about 20 amino acids of SEQ ID NO: 4.  
     
     
         38 . The purified antibody of  claim 37 , wherein said polypeptide comprising at least about twenty amino acids of the non-phosphorylated amino acid sequence of SEQ ID NO: 4 comprises amino acids 31-233 of LAT.  
     
     
         39 . The purified antibody of  claim 35 , wherein the polypeptide comprises SEQ ID NO: 4.  
     
     
         40 . The purified antibody of  claim 35 , wherein said polypeptide comprises the cytostolic tail of LAT.

         45 . The purified antibody of  claim 35 , wherein the antibody is a fully humanized antibody.

A purified antibody which is generated against a polypeptide comprising at least about five amino acids of the amino acid sequence of SEQ ID NO: 4, wherein the at least about five amino acids comprises a phosphotyrosine residue and wherein the antibody specifically binds to a phosphorylated LAT polypeptide comprising an amino acid sequence according to SEQ ID NO: 4.  
     
     
         47 . The purified antibody of  claim 46 , wherein the antibody is a polyclonal antibody.  
     
     
         48 . The purified antibody of  claim 47 , wherein the polypeptide comprising at least about five amino acids comprises SEQ ID NO: 4.  
     
     
         49 . The purified antibody of  claim 47 , wherein the polypeptide comprises at least about 20 amino acids of SEQ ID NO: 4.  
     
     
         50 . The purified antibody of  claim 49 , wherein said polypeptide comprising at least about twenty amino acids of the amino acid sequence of SEQ ID NO: 4 comprises amino acids 31-233 of LAT.  
     
     
         51 . The purified antibody of  claim 47 , wherein the polypeptide comprising from at least about five amino acids to at least about 20 amino acids of SEQ ID NO: 4.  
     
     
         52 . The purified antibody of  claim 47 , wherein said polypeptide comprises the cytostolic tail of LAT.

The purified antibody of  claim 47 , wherein the antibody immunoprecipitates a 40 kd protein from a lysate of C305 cells transfected with a LAT cDNA comprising a nucleic acid sequence according to SEQ ID NO: 1, but which does not immunoprecipitate the 40 kd protein from a lysate of non-transfected cells.
