Abstract

The present invention relates to a thermophilic arabinose isomerase and a method of manufacturing tagatose using the same, and more precisely, a gene encoding arabinose isomerase originating from the thermophilic Thermotoga neapolitana DSM 5068, a recombinant expression vector containing the gene, a method of preparing a food grade thermophilic arabinose isomerase from the recombinant GRAS (Generally Recognized As Safe) strain transformed with the said expression vector, and a method of preparing tagatose from galactose using the said enzyme.

1 . An arabinose isomerase expressed in the  Corynebacterium  sp. strain. 
     
     
         2 . The arabinose isomerase according to  claim 1 , wherein the arabinose isomerase originates from the  Thermotoga  sp.,  Thermus  sp. or  Sulforobus  sp. strain having an optimum reaction temperature of 70˜90° C. 
     
     
         3 . The arabinose isomerase according to  claim 2 , wherein the arabinose isomerase originates from  Thermotoga neapolitana  DSM 5068. 
     
     
         4 . The arabinose isomerase according to  claim 1 , wherein the arabinose isomerase contains the amino acid sequence represented by SEQ. ID. NO: 3. 
     
     
         5 . A method of preparing tagatose from galactose by using the arabinose isomerase of  claim 1 .
