Abstract

The present invention provides novel polynucleotides encoding HGPRBMY28 and HGPRBMY29 polypeptides, fragments and homologues thereof. The present invention also provides polynucleotides encoding splice variants of HGPRBMY29 polypeptides, HGPRBMY29v1 and HGPRBMY29v2. Also provided are vectors, host cells, antibodies, and recombinant and synthetic methods for producing said polypeptides. Also provided are vectors, host cells, antibodies, and recombinant and synthetic methods for producing said polypeptides. The invention further relates to diagnostic and therapeutic methods for applying these novel HGPRBMY28, HGPRBMY29, HGPRBMY29v1, and HGPRBMY29v2 polypeptides to the diagnosis, treatment, and/or prevention of various diseases and/or disorders related to these polypeptides. The invention further relates to screening methods for identifying agonists and antagonists of the polynucleotides and polypeptides of the present invention.

         21 . An isolated antibody or fragment thereof that binds to a polypeptide selected from the group consisting of:
 (a) the polypeptide comprising amino acids 1 to 560 of SEQ ID NO:43;   (b) the polypeptide comprising amino acids 2 to 560 of SEQ ID NO:43; and   (c) the polypeptide comprising amino acids 3 to 560 of SEQ ID NO:43.

         35 . The isolated antibody or fragment thereof of  claim 34 , wherein said detectable substance is selected from the group consisting of: a) an enzyme; b) a fluorescent label; and c) a radioisotope. 
     
     
         36 . The isolated antibody or fragment thereof of  claim 21 , which specifically binds to said polypeptide in an ELISA. 
     
     
         37 . The isolated antibody or fragment thereof of  claim 21 , which specifically binds to said polypeptide in a competitive-binding assay. 
     
     
         38 . The isolated antibody or fragment thereof of  claim 21 , which specifically binds to said polypeptide in a radioimmunoassay. 
     
     
         39 . The isolated antibody or fragment thereof of  claim 21 , which specifically binds to said polypeptide in a FACS. 
     
     
         40 . A kit for detecting a polypeptide comprising amino acids 3 to 560 of SEQ ID NO:43 comprising:
 the isolated antibody or fragment thereof of  claim 21 ; and   at least one component to detect binding of the isolated antibody to said polypeptide sequence.   
     
     
         41 . An isolated antibody obtained from an animal that has been immunized with a polypeptide according to  claim 21 , wherein said antibody binds to an antigenic epitope-bearing polypeptide fragment of said polypeptide. 
     
     
         42 . An isolated antibody or fragment thereof that binds to a polypeptide consisting of at least 473 contiguous amino acids of SEQ ID NO:43, wherein said polypeptide decreases the expression level of p21, p27, or IkBa in a cell in which said polypeptide is expressed 
     
     
         43 . An isolated antibody or fragment thereof that binds to a polypeptide consisting of at least 50 contiguous amino acids of SEQ ID NO:43. 
     
     
         44 . An isolated antibody or fragment thereof that binds to a polypeptide comprising a sequence provided as amino acids 3 to 560 of SEQ ID NO:43 containing a single amino acid substitution, wherein said polypeptide decreases the expression level of p21, p27, or IkBa in a cell in which said polypeptide is expressed.
