Abstract
X-ray fluorescence (XRF) spectrometry has been used for detecting binding events and measuring binding selectivities between chemicals and receptors. XRF may also be used for estimating the therapeutic index of a chemical, for estimating the binding selectivity of a chemical versus chemical analogs, for measuring post-translational modifications of proteins, and for drug manufacturing.

exact text as granted — not AI-modified1 . A method for estimating the binding affinity of a chemical having at least one heavy element to a receptor, comprising:
establishing a baseline X-ray fluorescence signal for a heavy element in the first receptor, the heavy element being present in a chemical to be tested for binding to the receptors; exposing the receptor to the chemical and allowing the chemical to bind to them to form a chemical-receptor complex; measuring the x-ray fluorescence signals due to the heavy element in the chemical-receptor complex; subtracting the baseline x-ray fluorescence signal of the receptor from the measured x-ray fluorescence signal of the of the chemical-receptor complex to obtain a net x-ray fluorescence signal; estimating the affinity of the chemical for the receptor by dividing the net x-ray fluorescence signal by the amount of receptor in the portion of the receptor to obtain a quotient of the chemical to the receptor.
2 . The method of claim 1 , wherein the receptor-chemical complex is deposited on a substrate having a thickness of less than 500 microns.
3 . The method of claim 1 , wherein the receptor-protein complex is deposited on a substrate having a thickness of between twenty (20) nanometers and twenty-five (25) microns.
4 . The method of claim 1 , wherein the receptor-chemical complex is deposited on a substrate comprising at least 10% of at least one of the following elements: beryllium, carbon, silicon, aluminum, iron, cobalt, and gold.
5 . The method of claim 1 , wherein the substrate is a focusing chip.
6 . The method of claim 5 , wherein the focusing chip is substantially free of at least one of the elements selected from the group of sulfur, fluorine, phosphorus, chlorine, bromine, iron, zinc, magnesium, calcium, titanium, scandium, and platinum.
7 . The method of claim 5 , wherein the focusing chip has one or more hydrophilic regions, wherein at least one of the hydrophilic regions has an area of between 75 square picometers and 3.6 square microns.
8 . The method of claim 1 , wherein the cross section of the excitation x-ray beam is between 25% and 250% of the area of the receptor-chemical complex.
9 . The method of claim 1 , wherein the protein-receptor complex is exposed to a solution prior to measurement, the solution comprising a buffer which is substantially free of at least one chemical element having an atomic number of greater than four, where that chemical element is present in the chemical.

The method of claim 1 , wherein the receptor-chemical complex is exposed to a solution prior to measurement, the solution comprising a buffer, the buffer being substantially free of at least one of the chemicals or functional groups selected from the group of dimethylsulfoxide, thiols, sulfate anion, sulfonate anions, chloride anion, bromide anion, fluoride anion, iodide anion, perchlorate anion, phosphate anion, and phosphonate anions.

The method of claim 1 , wherein the receptor-chemical complex is exposed to a solution prior to measurement, the solution comprising a buffer, and the buffer comprises one or more of the chemicals or functional groups selected from the group of amine, imine, nitrate anion, nitrite anion, ammonium cation, and iminium cation.
12 . The method of claim 1 , wherein the receptor-chemical complex is purified using gel filtration chromatography or dialysis prior to analysis.
13 . The method of claim 12 , wherein the receptor-chemical complex is purified using a chromatographic process which includes a vacuum or centrifugation step.
14 . The method of claim 1 , wherein the receptor-chemical complex is exposed to a solution prior to measurement, the solution comprising a matrix modifier.
15 . The method of claim 1 , wherein the x-ray fluorescence is measured using a chromium, palladium or silver x-ray source.
16 . The method of claim 1 , wherein the x-ray fluorescence is measured using an x-ray source having a K-alpha or L-alpha line at or above about 2.838 KeV and less than about 9.441 KeV.

The method of claim 1 , wherein the chemical comprises at least one of the chemicals selected from the list of abarelix, abciximab, acamprosate, acetazolamide, acetohexamide, acetophenazine, 19 . The method of claim 1 , wherein the unbound chemical is separated via gel filtration or another size exclusion technique, and an aliquot from each sample is spotted onto a substrate for XRF measurement.

A system for determining the binding constants of a chemical and a receptor, comprising a process for equilibrating the chemical with the protein by incubating the chemical and protein in solution for sufficient time to ensure that chemical equilibrium is obtained; separating the unbound chemical in less than 5 minutes at 37 degrees Celsius, and determining the x-ray fluorescence signal of the chemical, and converting the x-ray fluorescence signal to a concentration.

An apparatus for measuring protein characteristics comprising an x-ray fluorescence spectrometer, a protein, and a security system comprising at least one of the security controls selected from the list of a biometric analysis system, a closed system, digital signatures, digital signatures configured so that no two individuals have the same combination of identification code and password.
24 . A method for analyzing protein purity, comprising exposing a protein sample to x-rays, measuring any x-ray fluorescence, converting the x-ray fluorescence signals of the elements measured into molar ratios, and comparing the molar ratios of the elements measured to a control value.
25 . An apparatus comprising an X-ray source, and x-ray detector, a protein sample deposited on a substrate, the substrate having a thickness of between 20 nanometers and 25 microns, and a focusing optic disposed between the protein and the x-ray source.
26 . The apparatus of claim 25 , wherein the x-ray source emits a beam of x-rays, and wherein the beam of x-rays irradiates an area of between one square nanometer and 3 square microns.
27 . The apparatus of claim 25 , wherein the x-ray source emits a beam of x-rays and wherein the beam of x-rays irradiates a cross-sectional area that is between 25% and 250% of the area of the protein sample.
28 . The source of claim 25 , further comprising a collimator disposed between the source and the protein sample.
29 . An X-ray microscope for measuring a sample comprising:
an X-ray source disposed so as to be able to expose the sample to X-rays; a detector disposed so as to be able to detect the X-ray fluorescence of the sample; a recording means for making a record of the X-ray fluorescence; and a record-control means.

exposing the protein composition to X-rays; measuring the amount of sulfur in the protein composition by measuring the X-ray fluorescence in the sulfur spectrum; measuring the amount of a non-sulfur element in the protein composition by measuring the X-ray fluorescence in the non-sulfur-element spectrum; and calculating the relative ratio of the amount of the sulfur and the amount of the non-sulfur element, thus monitoring the protein composition.
48 . A method for monitoring the purity of a protein composition, the method comprising the steps of:
exposing the protein composition to X-rays; exposing the protein composition to light having a wavelength of about 260 nm; measuring the amount of sulfur in the protein composition by measuring the X-ray fluorescence in the sulfur spectrum; measuring the amount of protein in the protein composition by measuring the absorbance of the light having a wavelength of about 260 nm; and calculating the relative ratio of the amount of sulfur a the amount of protein.Cited by (0)
