4-amino-2,3-disubstituted thieno [2,3,d]pyrimidines and pharmaceutical compositions thereof

Abstract

Furo- and thienopyrimidine derivatives, which are useful as TIE-2 and/or VEGFR-2 inhibitors are described herein. The described invention also includes methods of making such furo- and thienopyrimidine derivatives as well as methods of using the same in the treatment of hyperproliferative diseases.

 A is hydrogen, halo, C 1 -C 6  alkyl, aryl, heteroaryl, aryl or heteroaryl substituted with at least one independently selected R 3  group, heterocyclyl, —RR 3 , —C(O)OR 4 , —C(O)NR 5 R 6 , —C(O)R 4 ; 
 D is hydrogen, halo, C 1 -C 6  alkyl, aryl, heteroaryl, aryl or heteroaryl substituted with at least one independently selected R 3  group, heterocyclyl, —RR 3 , —C(O)OR 4 , —C(O)NR 5 R 6 , or —C(O)R 4 ; 
 R is C 1 -C 6  alkylene, C 3 -C 7  cycloalkylene, C 1 -C 6  alkenylene, or C 1 -C 6  alkynylene; 
 R 1  is hydrogen, C 1 -C 6  alkyl, C 1 -C 6  alkoxy, —SR 4 , —S(O) 2 R 4 , —NR 7 R 7 , —NR′NR′″R″″, —N(H)RR 3 , —C(O)OR 7 , or —C(O)NR 7 R 7 ; 
 R 2  is hydrogen, —OH, —NR 7 R 7  or ═NH; 
 R 3  is halo, C 1 -C 6  alkyl, C 1 -C 6  haloalkyl, C 1 -C 6  alkoxy, C 3 -C 7  cycloalkoxy, C 1 -C 6  haloalkoxy, aryl, aralkyl, aryloxy, heteroaryl, heterocyclyl, —CN, —NHC(O)R 4 , —N(R 3 )HC(O)R 4 , —NHC(S)R 4 , —NR 5 R 6 , —RNR 5 R 6 , —SR 4 , —S(O) 2 R 4 , —RC(O)OR 4 , —C(O)OR 4 , —C(O)R 4 , —C(O)NR 5 R 6 , —NHS(O) 2 R 4 , —N(S(O) 2 R 4 )S(O) 2 R 4 , —S(O) 2 NR 5 R 6 , or —NHC(═NH)R 4 ; 
 R 4  is hydrogen, C 1 -C 6  alkyl, aryl, heteroaryl, heterocyclyl, —RR 3 , —NR′″R″″, or —NR′NR′″R″″; 
 R 5  is hydrogen, C 1 -C 6  alkyl, C 3 -C 7  cycloalkyl, cyanoalkyl, —R′R″, aryl, aralkyl, heteroaryl, —NHC(O)OR′″, —R′NHC(O)OR′″, —R′NHC(O)NR′″R″″, or —R′C(O)OR′″; 
 R 6  is hydrogen, C 1 -C 6  alkyl, C 3 -C 7  cycloalkyl, cyanoalkyl, —R′R″, aryl, aralkyl, heteroaryl, —C(O)OR′″, or —R′C(O)NR′″R″″; 
 R 7  is hydrogen, C 1 -C 6  alkyl, aryl, or —C(O)OR′″; 
 R 8  is C 1 -C 3  alkyl; 
 R′ is C 1 -C 3  alkylene; 
 R″ is heteroalkyl or NR′″R″″; 
 R′″ is hydrogen, C 1 -C 6  alkyl, aryl, aralkyl, heteroaryl, or C 3 -C 7  cycloalkyl; 
 R″″ is hydrogen, C 1 -C 6  alkyl, aryl, heteroaryl, or C 3 -C 7  cycloalkyl.

       4 . A compound as claimed in  claim 1 , wherein A is aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group. 
   
   
       5 . A compound as claimed in  claim 1 , wherein A is aryl substituted with at least one independently selected R 3  group. 
   
   
       6 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with at least one independently selected R 3  group. 
   
   
       7 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with the group —NHC(O)R 4 , wherein R 4  is the group NR′″R″″. 
   
   
       8 . A compound as claimed in  claim 1 , wherein D is hydrogen, halo, aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group. 
   
   
       9 . A compound as claimed in  claim 1 , wherein D is hydrogen, halo, or aryl substituted with at least one independently selected R 3  group. 
   
   
       10 . A compound as claimed in  claim 1 , wherein D is aryl substituted with at least one independently selected R 3  group. 
   
   
       11 . A compound as claimed in  claim 1 , wherein D is hydrogen or halo. 
   
   
       12 . A compound as claimed in  claim 1 , wherein A is aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group and D is hydrogen, halo, aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group. 
   
   
       13 . A compound as claimed in  claim 1 , wherein A is aryl substituted with at least one independently selected R 3  group and D is hydrogen, halo, or aryl substituted with at least one independently selected R 3  group. 
   
   
       14 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with at least one independently selected R 3  group and D is hydrogen or halo. 
   
   
       15 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with at least one independently selected R 3  group and D is aryl substituted with at least one independently selected R 3  group. 
   
   
       16 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with the group —NHC(O)R 4 , wherein R 4  is the group NR′″R″″ and D is aryl substituted with at least one independently selected R 3  group. 
   
   
       17 . A compound as claimed in  claim 1 , wherein A is phenyl substituted with the group —NHC(O)R 4 , wherein R 4  is the group NR′″R″″ and D is hydrogen or halo. 
   
   
       18 . A compound as claimed in  claim 1 , wherein R 1  is hydrogen, methyl, —C(O)NH 2 , —NH 2 , —NHCH 2 CH 2 NR′″R″″. 
   
   
       19 . A compound as claimed in  claim 1 , wherein R 1  is hydrogen, methyl or —NH 2 . 
   
   
       20 . A compound as claimed in  claim 1 , wherein R 1  is hydrogen. 
   
   
       21 . A compound as claimed in  claim 1 , wherein R 2  is —NR 7 R 7 . 
   
   
       22 . A compound as claimed in  claim 1 , wherein R 2  is —NR 7 R 7  wherein each R 7  groups is independently selected from hydrogen or C 1 -C 6  alkyl. 
   
   
       23 . A compound as claimed in  claim 1 , wherein R 2  is —NH 2 . 
   
   
       24 . A compound as claimed in  claim 1 , wherein X is O, A is aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group, D is hydrogen, halo, aryl, heteroaryl, or aryl or heteroaryl substituted with at least one independently selected R 3  group, R 1  is hydrogen, methyl, —C(O)NH 2 , —NH 2 , or —NHCH 2 CH 2 NR′″R″″, and R 2  is —NR 7 R 7 .

A compound as claimed in  claim 1 , wherein X is O, A is aryl substituted with at least one independently selected R 3  group, D is hydrogen, halo, or aryl substituted with at least one independently selected R 3  group, R 1  is hydrogen, methyl, —C(O)NH 2 , —NH 2 , or —NHCH 2 CH 2 NR′″R″″, and R 2  is —NR 7 R 7 .

A compound as claimed in  claim 1 , wherein X is O, A is phenyl substituted with at least one independently selected R 3  group, D is hydrogen or halo, R 1  is hydrogen, methyl or —NH 2 , and R 2  is —NR 7 R 7  wherein the R 7  groups are selected from hydrogen or C 1 -C 6  alkyl. 
   
   
       27 . A compound as claimed in  claim 1 , wherein X is O, A is phenyl substituted with at least one independently selected R 3  group, D is aryl substituted with at least one independently selected R 3  group, R 1  is hydrogen or methyl, and R 2  is —NR 7 R 7  wherein the R 7  groups are selected from hydrogen or C 1 -C 6  alkyl. 
   
   
       28 . A compound as claimed in  claim 1 , wherein X is O, A is phenyl substituted with the group —NHC(O)R 4 , wherein R 4  is the group NR′″R″″, D is aryl substituted with at least one independently selected R 3  group, R 1  is hydrogen, and R 2  is —NH 2 . 
   
   
       30 . A compound as claimed in  claim 1 , wherein X is O, A is phenyl substituted with the group —NHC(O)R 4 , wherein R 4  is the group NR′″R″″, D is hydrogen or halo, R 1  is hydrogen, and R 2  is —NH 2 .

       34 . A pharmaceutical composition, comprising: a therapeutically effective amount of a compound as claimed in  claim 1  and one or more of pharmaceutically acceptable carriers, diluents and excipients. 
   
   
       35 . The pharmaceutical composition of  claim 34 , further comprising at least one additional anti-neoplastic agent. 
   
   
       36 . The pharmaceutical composition of  claim 34 , further comprising an additional agent which inhibits angiogenesis.
