Abstract
The application describes neutralizing chimeric and humanized anti-human IL-9 antibodies, and the use thereof to identify neutralizing epitopes on human IL-9 and as medicaments to prevent and treat asthma, bronchial hyperresponsiveness, atopic allergy, and other related disorders. Particularly disclosed are recombinant antibodies derived from three murine anti-human IL-9 antibodies identified infra as MH9A3, MH9D1, and MH9L1.

11 . A humanized antibody which specifically binds to human IL-9 comprising at least one CDR with an amino acid sequence with at least 85% sequence identity to any CDR from antibody MH9A3.
12 . The humanized antibody of claim 11 , wherein the CDR is selected from the group consisting of CDR1, CDR2, and CDR3.
13 . The humanized antibody of claim 12 , wherein the CDR is in the heavy chain.
14 . The humanized antibody of claim 13 , wherein the amino acid sequence of CDR1 comprises amino acids 26 to 35 of SEQ ID NO: 3.
15 . The humanized antibody of claim 13 , wherein the amino acid sequence of CDR1 consists of amino acids 26 to 35 of SEQ ID NO: 3.
16 . The humanized antibody of claim 13 , wherein the amino acid sequence of CDR2 has at least 90% sequence identity to CDR2 (amino acids 50 to 66 of SEQ ID NO: 3) of MH9A3.
17 . The humanized antibody of claim 16 , wherein the amino acid sequence of CDR2 comprises amino acids 50 to 66 of SEQ ID NO: 3.
18 . The humanized antibody of claim 16 , wherein the amino acid sequence of CDR2 consists of amino acids 50 to 66 of SEQ ID NO: 3.
19 . The humanized antibody of claim 13 , wherein the amino acid sequence of CDR3 has at least 90% sequence identity to CDR3 (amino acids 99 to 111 of SEQ ID NO: 3) of MH9A3.
20 . The humanized antibody of claim 19 , wherein the amino acid sequence of CDR2 comprises amino acids 99 to 111 of SEQ ID NO: 3.
21 . The humanized antibody of claim 19 , wherein the amino acid sequence of CDR2 consists of amino acids 99 to 111 of SEQ ID NO: 3.
22 . The humanized antibody of claim 12 , wherein the CDR is in the light chain.

i. a variable heavy chain containing a CDR2 and CDR3 with an amino acid sequence with at least 90% sequence identity to CDR2 (amino acids 50 to 66 of SEQ ID NO: 3) and CDR3 (amino acids 50 to 66 of SEQ ID NO: 3) of MH9A3 heavy chain; and ii. a variable light chain containing a CDR1 with an amino acid sequence with at least 90% sequence identity to CDR1 (amino acids 24 to 34 of SEQ ID NO: 5) of MH9A3 light chain.Cited by (0)
