Abstract

The present invention relates to a novel human protein called Human G-protein Chemokine Receptor (CCR5) HDGNR10, and isolated polynucleotides encoding this protein. The invention is also directed to human antibodies that bind Human G-protein Chemokine Receptor (CCR5) HDGNR10 and to polynucleotides encoding those antibodies. Also provided are vectors, host cells, antibodies, and recombinant methods for producing Human G-protein Chemokine Receptor (CCR5) HDGNR10 and human anti-Human G-protein Chemokine Receptor (CCR5) HDGNR10 antibodies. The invention further relates to diagnostic and therapeutic methods useful for diagnosing and treating diseases, disorders, and/or conditions related to this novel human protein and these novel human antibodies.

Claims

An isolated antibody or fragment there of comprising a VH domain and a VL domain wherein the amino acid sequence of said VH domain and said VL domain are at least 85% identical to the amino acid sequence of the VH and the VL domain, respectively, of the antibody expressed by a hybridoma cell line selected from the group consisting of:
 (a) XF3.5F1;   (b) XFl 1.1F8;   (c) XF3.6A2;   (d) XF3.10B8;   (e) XF22.3C9.6;   (f) XF22.9E6;   (g) XF27/28.7D5;   (h) XF27/28.18B5;   (i) XF27/28.25G10;   j) XF27/28.36A12; and   (k) XF27/28.36F11.   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         2 . An isolated antibody or fragment thereof comprising:
 (a) an amino acid sequence that is at least 85% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 85% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         3 . The antibody or fragment thereof of  claim 2  comprising:
 (a) an amino acid sequence that is at least 90% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 90% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         4 . The antibody or fragment thereof of  claim 2  comprising:
 (a) an amino acid sequence that is at least 95% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 95% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         5 . The antibody or fragment thereof of  claim 2  comprising:
 (a) the amino acid sequence of the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) the amino acid sequence of the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         6 . An isolated cell that produces the antibody or fragment thereof of  claim 2 . 
     
     
         7 . The XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053. 
     
     
         8 . The antibody produced by the XF27/28.36F11 hybridoma cell line of  claim 7 . 
     
     
         9 . An isolated human antibody or fragment thereof that binds the same epitope on a CCR5 polypeptide as the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053. 
     
     
         10 . An isolated human antibody or fragment thereof that competitively inhibits the binding of the antibody produced by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053. 
     
     
         11 . An isolated human antibody or fragment thereof comprising:
 (a) an amino acid sequence that is at least 85% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 85% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         12 . The antibody or fragment thereof of  claim 11  comprising:
 (a) an amino acid sequence that is at least 90% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 90% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         13 . The antibody or fragment thereof of  claim 11  comprising:
 (a) an amino acid sequence that is at least 95% identical to the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) an amino acid sequence that is at least 95% identical to the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         14 . The antibody or fragment thereof of  claim 11  comprising:
 (a) the amino acid sequence of the VH domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053; and   (b) the amino acid sequence of the VL domain of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053;   
       wherein said antibody or fragment thereof specifically binds a CCR5 polypeptide. 
     
     
         15 . An isolated antibody or fragment thereof comprising the amino acid sequence of the VHCDR1, VHCDR2, VHCDR3, VLCDR1, VLCDR2, and VLCDR3 of the antibody expressed by the XF27/28.36F11 hybridoma cell line deposited under ATCC Deposit Accession Number PTA-4053, wherein said antibody or fragment thereof specifically binds CCR5.
