Abstract

Novel polypeptide combinations, polynucleotides encoding the polypeptides, and related compositions and methods are disclosed for zcytor17-containing multimeric or heterodimer cytokine receptors that may be used as novel cytokine antagonists, and within methods for detecting ligands that stimulate the proliferation and/or development of hematopoietic, lymphoid and myeloid cells in vitro and in vivo. The present invention also includes methods for producing the multimeric or heterodimeric cytokine receptor, uses therefor and antibodies thereto.

A monoclonal antibody or antibody fragment that specifically binds to a multimeric or heterodimeric cytokine receptor comprising a first cytokine receptor consisting of an amino acid sequence having at least 95% sequence identity with amino acid residues 20-519 of SEQ ID NO:111 and a second cytokine receptor comprising amino acid residues 28-739 of SEQ ID NO:7. 
     
     
         2 . The monoclonal antibody or antibody fragment of  claim 1  wherein the monoclonal antibody or antibody fragment is selected from the group consisting of a murine monoclonal antibody, a human monoclonal antibody, a humanized antibody derived from a murine monoclonal antibody, a chimeric antibody, Fab fragment, F(ab′)2 fragment, and a single chain antibody. 
     
     
         3 . The monoclonal antibody or antibody fragment of  claim 1  wherein the monoclonal antibody or antibody fragment further comprises a radionuclide, enzyme, substrate, cofactor, fluorescent marker, chemiluminescent marker, peptide tag, magnetic particle, drug, or toxin. 
     
     
         4 . A composition comprising the monoclonal antibody or antibody fragment of  claim 1  and a pharmaceutically acceptable vehicle. 
     
     
         5 . A monoclonal antibody that specifically binds to a multimeric or heterodimeric cytokine receptor comprising a first cytokine receptor and a second cytokine receptor:
 (a) said first cytokine receptor comprises an amino acid sequence that is at least 95% identical to amino acid residues 20-519 of SEQ ID NO:111 and includes a cytokine binding domain consisting essentially of amino acid residues 20-227 of SEQ ID NO:111; and   (b) said second cytokine receptor comprises amino acid residues 28-739 of SEQ ID NO:7.   
     
     
         6 . The monoclonal antibody of  claim 5  wherein said monoclonal antibody is selected from the group consisting of, a murine monoclonal antibody, a human monoclonal antibody, a humanized antibody derived from a murine monoclonal antibody, an antibody fragment, a chimeric antibody, Fab fragment, F(ab′)2 fragment, a single chain antibody and combinations thereof. 
     
     
         7 . The monoclonal antibody of  claim 5  wherein said monoclonal antibody further comprises a radionuclide, enzyme, substrate, cofactor, fluorescent marker, chemiluminescent marker, peptide tag, magnetic particle, drug, or toxin. 
     
     
         8 . A composition comprising said monoclonal antibody of  claim 5  and a pharmaceutically acceptable vehicle. 
     
     
         9 . A monoclonal antibody that specifically binds to a multimeric or heterodimeric cytokine receptor comprising a first cytokine receptor consisting of an amino acid sequence having at least 95% sequence identity with amino acid residues 20-519 of SEQ ID NO:111 and a second cytokine receptor comprising amino acid residues 28-739 of SEQ ID NO:7, wherein said antibody binding to said multimeric or heterodimeric receptor will antagonize a zcytor17lig mediated activity of said multimeric or heterodimeric receptor. 
     
     
         10 . The monoclonal antibody of  claim 9  wherein said monoclonal antibody is selected from the group consisting of a murine monoclonal antibody, a human monoclonal antibody, a humanized antibody derived from a murine monoclonal antibody, an antibody fragment, a chimeric antibody, Fab fragment, F(ab′)2 fragment, a single chain antibody and combinations thereof. 
     
     
         11 . The monoclonal antibody of  claim 9  wherein said monoclonal antibody further comprises a radionuclide, enzyme, substrate, cofactor, fluorescent marker, chemiluminescent marker, peptide tag, magnetic particle, drug, or toxin. 
     
     
         12 . A composition comprising said antibody of  claim 9  and a pharmaceutically acceptable vehicle.
