Abstract

The invention relates to Brachyspira pilosicoli 72 kDa outer-membrane protein (Bpmp-72) and its amino acid and nucleotide sequence. The invention also relates to the uses of these sequences in prophylactic (vaccination) and therapeutic treatment of infections with Brachyspira pilosicoli (internal spirochaetosis).

         27 . An isolated polypeptide comprising an amino acid sequence set forth in SEQ ID NO:2 or analogue thereof having at least 60% homology thereto, wherein the isolated polypeptide is capable of eliciting an immune response to  Brachyspira pilosicoli , and wherein the isolated polypeptide is capable of eliciting an antibody that specifically binds to the 34 kDa C-terminal portion of the Bpmp-72 outer membrane protein that consists of the amino acid sequence set forth in SEQ ID NO:2. 
     
     
         28 . The isolated polypeptide of  claim 27 , wherein the analogue has at least 70% homology to the amino acid sequence set forth in SEQ ID NO:2. 
     
     
         29 . The isolated polypeptide of  claim 27 , wherein the analogue has at least 80% homology to the amino acid sequence set forth in SEQ ID NO:2. 
     
     
         30 . The isolated polypeptide of  claim 27 , wherein the analogue has at least 90% homology to the amino acid sequence set forth in SEQ ID NO:2. 
     
     
         31 . The isolated polypeptide of  claim 27 , wherein the analogue has at least 95% homology to the amino acid sequence set forth in SEQ ID NO:2. 
     
     
         32 . The isolated polypeptide of  claim 27 , wherein the analogue has at least 98% homology to the amino acid sequence set forth in SEQ ID NO:2.

The isolated polypeptide of  claim 27 , wherein the analogue has an amino acid sequence selected from the group consisting of SEQ ID NO:3, SEQ ID NO:4, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:7, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO:10, SEQ ID NO:1, SEQ ID NO: 12, SEQ ID NO:13, SEQ ID NO: 14, SEQ ID NO:15, SEQ ID NO:16, SEQ ID NO: 17, SEQ ID NO:8, SEQ ID NO:19, SEQ ID NO:20, SEQ ID NO:21, and SEQ ID NO:22.

         38 . A method of immunizing a subject against infection with Brachyspira pilosicoli comprising administering the vaccine composition of  claim 37  to a subject in need thereof.
