Abstract
Compounds useful for treating cellular proliferative diseases and disorders by modulating the activity of one or more mitotic kinesins are disclosed.

and pharmaceutically acceptable salts, solvates, chelates, non-covalent complexes, prodrugs, and mixtures thereof, wherein
R 1 is chosen from 3-halo-4-((R)-1,1,1-trifluoropropan-2-yloxy)phenyl, 3-cyano-4-((R)-1,1,1-trifluoropropan-2-yloxy)phenyl, 3-halo-4-isopropylamino-phenyl, 3-cyano-4-isopropylamino-phenyl, 3-halo-4-((R)-1,1,1-trifluoropropan-2-ylamino)phenyl, and 3-cyano-4-((R)-1,1,1-trifluoropropan-2-ylamino)phenyl;
X is chosen from —CO and —SO 2 —;
R 2 is chosen from hydrogen and optionally substituted lower alkyl;
W is chosen from —CR 4 —, —CH 2 CR 4 —, and N;
R 3 is chosen from —CO—R 7 , hydrogen, optionally substituted alkyl, optionally substituted heterocycloalkyl, optionally substituted heteroaryl, cyano, sulfonyl, and optionally substituted aryl;
R 4 is chosen from hydrogen and optionally substituted alkyl;
R 5 is chosen from hydrogen, hydroxy, optionally substituted amino, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl;optionally substituted heteroaryl, and optionally substituted lower alkyl;
R 6 is chosen from hydrogen, optionally substituted alkyl, optionally substituted alkoxy, optionally substituted aryloxy, optionally substituted heteraryloxy, optionally substituted alkoxycarbonyl-, aminocarbonyl-, optionally substituted aryl, optionally substituted heteroaryl, optionally substituted cycloalkyl, and optionally substituted heterocycloalkyl; and
R 7 is chosen from optionally substituted lower alkyl, optionally substituted aryl, hydroxy, optionally substituted amino, optionally substituted aralkoxy, and optionally substituted alkoxy;
provided that if W is N, then R 5 is not hydroxy or optionally substituted amino, and R 6 is not optionally substituted alkoxy, optionally substituted aralkoxy, optionally substituted heteroaralkoxy, or optionally substituted amino.
2 . At least one chemical entity of claim 1 wherein X is —CO—.
3 . At last one chemical entity of claim 1 or 2 wherein W is —CR 4 —.
4 . At least one chemical entity of any one of claims 1 to 3 wherein R 4 is hydrogen.
5 . At least one chemical entity of any one of claims 1 to 4 wherein R 5 is chosen from hydrogen, hydroxy, and optionally substituted lower alkyl.
6 . At least one chemical entity of claim 5 wherein R 5 is hydrogen.
7 . At least one chemical entity of any one of claims 1 to 6 wherein R 3 is chosen from —CO—R 7 ; hydrogen; optionally substituted lower alkyl; cyano; sulfonyl; optionally substituted aryl; optionally substituted heteroaryl, and optionally substituted heterocycloalkyl.
8 . At least one chemical entity of claim 7 wherein R 3 is optionally substituted lower alkyl or optionally substituted heteroaryl.
9 . At least one chemical entity of claim 8 wherein R 3 is chosen from lower alkyl that is optionally substituted with a hydroxy or a phosphate or acyl ester thereof, lower alkyl that is optionally substituted with a lower alkoxy, lower alkyl that is optionally substituted with an optionally substituted amino group, and lower alkyl that is optionally substituted with CO—R 7 where R 7 is chosen from hydroxy and optionally substituted amino.
10 . At least one chemical entity of claim 9 wherein R 3 is chosen from lower alkyl that is optionally substituted with a hydroxy or a phosphate or acyl ester thereof and lower alkyl that is optionally substituted with an optionally substituted amino group.
11 . At least one chemical entity of claim 10 wherein R 3 is 2-hydroxyethyl.
12 . At least one chemical entity of any one of claims 1 to 11 wherein R 6 is chosen from optionally substituted aryl, optionally substituted heteroaryl, optionally substituted heterocycloalkyl, optionally substituted cycloalkyl, and optionally substituted alkyl.

At least one chemical entity of claim 12 wherein R 6 is phenyl substituted with one or two of the following substituents: optionally substituted heteroaryl, optionally substituted amino, aralkoxy, halo, hydroxymethyl-, hydroxy, cyano, alkoxy, phenyl, phenoxy, methylenedioxy, ethylenedioxy, sulfonyl, aminocarbonyl, carboxy, alkoxycarbonyl, nitro, heteroaralkoxy, aralkoxy, and optionally substituted heterocycloalkyl.
14 . At least one chemical entity of claim 12 wherein R 6 is phenyl substituted with R 14 at the para position and further substituted with a group R 15 wherein
R 14 is chosen from optionally substituted heterocycloalkyl and optionally substituted heteroaryl; and R 15 is chosen from hydrogen, halo, hydroxy, and lower alkyl.
15 . At least one chemical entity of claim 14 , wherein R 14 is chosen from
7,8-dihydro-imidazo[1,2-c][1,3]oxazin-2-yl, 3a,7a-dihydro-1H-benzoimidazol-2-yl, imidazo[2,1-b]oxazol-6-yl, oxazol-4-yl, 5,6,7,8-tetrahydro-imidazo[1,2-a]pyridin-2-yl, 1H-[1,2,4]triazol-3-yl, 2,3-dihydro-imidazol-4-yl, 1H-imidazol-2-yl, imidazo[1,2-a]pyridin-2-yl, thiazol-2-yl, thiazol-4-yl, pyrazol-3-yl, and 1H-imidazol-4-yl,
each of which is optionally substituted with one, two, or three groups chosen from optionally substituted lower alkyl, halo, acyl, sulfonyl, cyano, nitro, optionally substituted amino, and optionally substituted heteroaryl.
16 . At least one chemical entity of claim 15 , wherein R 14 is chosen from
1H-imidazol-2-yl, imidazo[1,2-a]pyridin-2-yl; and 1H-imidazol-4-yl,
each of which is optionally substituted with one or two groups chosen from optionally substituted lower alkyl, halo, and acyl.
17 . At least one chemical entity of any one of claims 14 to 16 wherein R 15 is hydrogen.
18 . At least one chemical entity of claim 1 wherein W is N, R 5 is hydrogen; and R 6 is phenyl substituted at the para position with a group R 16 and further substituted with a group R 17 wherein
R 16 is chosen from halo, optionally substituted aryl, optionally substituted heteroaryl, optionally substituted alkoxy, and optionally substituted amino; and R 17 is chosen from hydrogen, lower alkoxy, halo, and lower alkyl.
19 . At least one chemical entity of claim 18 wherein R 16 is chosen from optionally substituted phenyl, optionally substituted heteroaryl, optionally substituted lower alkoxy, and optionally substituted amino.
20 . At least one chemical entity of claim 19 wherein R 16 is chosen from phenyl, optionally substituted benzyloxy, halo, optionally substituted 1H-imidazol-4-yl, optionally substituted 1H-benzo[d]imidazol-2-yl, and optionally substituted cyclohexylmethoxy.
21 . At least one chemical entity of any one of claims 18 to 20 wherein R 17 is hydrogen.
22 . At least one chemical entity of claim 1 wherein R 5 is hydrogen, W is CH, and R 3 is —CH 2 NH(CO)R 9 wherein
R 9 is chosen from optionally substituted alkoxy, optionally substituted cycloalkoxy, optionally substituted arylalkoxy, optionally substituted amino and optionally substituted lower alkyl.
23 . At least one chemical entity of claim 22 wherein R 9 is chosen from lower alkyl substituted with a group chosen from hydroxy and optionally substituted amino.
24 . At least one chemical entity of claim 23 wherein R 9 is lower alkyl substituted with a group chosen from hydroxy, amino, azetidino, N-methylamino, and N,N-dimethylamino.
25 . At least one chemical entity of any one of claims 1 to 24 wherein R 2 is hydrogen.

28 . A composition according to claim 27 , wherein said composition further comprises a chemotherapeutic agent other than a compound of Formula I.
29 . A composition according to claim 28 , wherein said composition further comprises a taxane, a vinca alkaloid, or a topoisomerase I inhibitor.
30 . A method of modulating CENP-E kinesin activity which comprises contacting said kinesin with an effective amount of at least one chemical entity of any one of claims 1 to 26 .
31 . A method of inhibiting CENP-E which comprises contacting said kinesin with an effective amount of at least one chemical entity of any one of claims 1 to 26 .
32 . A method for the treatment of a cellular proliferative disease comprising administering to a subject in need thereof at least one chemical entity of any one of claims 1 to 26 .
33 . A method for the treatment of a cellular proliferative disease comprising administering to a subject in need thereof a composition according to any one of claims 27 to 29 .
