Abstract
A novel class of compounds of the general formula (I), their use in therapy, pharmaceutical compositions comprising the compounds, as well as their use in the manufacture of medicaments are described. The present compounds modulate the activity of 11β-hydroxysteroid dehydrogenase type 1 (11βHSD1) and are accordingly useful in the treatment of diseases in which such a modulation is beneficial, e.g. the metabolic syndrome.

A pharmaceutical composition which is for oral, nasal, buccal, transdermal, pulmonal or parenteral administration, comprising an active ingredient together with one or more pharmaceutically acceptable carriers or excipients, wherein said active ingredient is at least one compound according to formula (I):

R 1 and R 2 together with the nitrogen to which they are attached, are forming a saturated or partially saturated bicyclic or tricyclic ring system containing from 4 to 12 carbon atoms and from 0 to 2 additional heteroatoms selected from nitrogen or oxygen, the ring system optionally being substituted with at least one of R 5 , R 5 oxy-, R 6 , hydroxy, oxo, cyano, C 1 -C 6 alkyl, aryl, hetaryl, arylC 1 -C 6 alkyl, hetarylC 1 -C 6 alkyl or C 1 -C 6 alkyloxyC 1 -C 6 alkyl, wherein the alkyl and aryl groups independently are optionally substituted with one ore more of R 7 ;
R 3 is C 1 -C 6 alkyl, —NR 8 R 9 , —C(═O)NR 8 R 9 or —OR 10 , wherein the alkyl group is optionally substituted with one or more of R 11 ;
R 4 is hydrogen, halo, hydroxy, cyano, trihalomethyl or C 1 -C 6 alkyl;

R 12 and R 13 together with the nitrogen to which they are attached, are forming a saturated or partially saturated cyclic, bicyclic or tricyclic ring system containing from 4 to 12 carbon atoms and from 0 to 2 additional heteroatoms selected from nitrogen or oxygen, the ring system optionally being substituted with at least one R 5 , R 5 oxy-, R 6 , halo, cyano, hydroxy, oxo, C 1 -C 8 alkyl, aryl, hetaryl, arylC 1 -C 6 alkyl, hetarylC 1 -C 6 alkyl, C 1 -C 6 alkyloxyC 1 -C 6 alkyl, NR 12 R 13 carbonylC 1 -C 6 alkyl, NR 12 R 13 C 1 -C 6 alkylcarbonyl-, R 14 C 1 -C 6 alkylcarbonyl- or —COR 15 ;

2 . A pharmaceutical composition according to claim 1 wherein X and Y are carbon.
3 . A pharmaceutical composition according to claim 1 wherein R 3 is —NR 8 R 9 , —C(═O)NR 8 R 9 or —OR 10 .
4 . A pharmaceutical composition according to claim 3 wherein R 3 is —NR 8 R 9 .
5 . A pharmaceutical composition according to claim 1 wherein R 1 and R 2 together with the nitrogen to which they are attached, are forming a saturated or partially saturated bicyclic or tricyclic ring system containing from 4 to 12 carbon atoms and from 0 to 2 additional heteroatoms selected from nitrogen or oxygen, the ring system optionally being substituted with at least one of R 6 , hydroxyl or C 1 -C 6 alkyl, wherein the alkyl groups independently are optionally substituted with one ore more of R 7 .
6 . A pharmaceutical composition according to claim 1 wherein:
a) R 4 is hydrogen or halo; b) R 5 is C 3 -C 10 cycloalkylcarbonyl- or C 3 -C 10 cycloalkylC 1 -C 6 alkylcarbonyl-; c) R 6 is C 1 -C 6 alkyloxy-; d) R 8 is hydrogen, C 1 -C 6 alkyl, C 3 -C 10 cycloalkyl or C 3 -C 10 cycloalkylC 1 -C 6 alkyl; e) R 9 is C 1 -C 6 alkyl, C 3 -C 10 cyclo-alkyl, C 3 -C 10 cycloalkylC 1 -C 6 alkyl, C 3 -C 10 cycloalkylcarbonyl-, C 3 -C 10 hetcycloalkylcarbonyl-, arylcarbonyl-, hetarylcarbonyl-, C 1 -C 6 alkyloxyC 1 -C 6 alkyl, NR 12 R 13 carbonylC 1 -C 6 alkyl-, R 14 C 1 -C 6 alkylcarbonyl-, —COR 15 , arylC 1 -C 6 alkylS(O) n —, arylC 1 -C 6 alkyl or hetarylC 1 -C 6 alkyl, wherein the alkyl, aryl and hetaryl groups independently are optionally substituted with one or more R 11 ; f) R 10 is C 1 -C 6 alkyl, wherein the alkyl group is optionally substituted with one or more R 11 ; g) R 11 is R 5 , R 6 , halo, hydroxy, C 1 -C 8 alkyloxy, oxo, cyano, —COR 15 , C 1 -C 8 alkyl or trihalomethyl; or h) R 14 is C 1 -C 6 alkyloxy, C 3 -C 10 cycloalkyloxy-, C 3 -C 10 cycloalkylC 1 -C 6 alkyloxy-, C 3 -C 10 hetcycloalkyloxy-, arylC 1 -C 6 alkyloxy, hetarylC 1 -C 6 alkyloxy, —NR 12 R 13 , or —COR 15 , wherein the alkyl, cycloalkyl, hetcycloalkyl, aryl and hetaryl groups independently are optionally substituted with one or more of R 20 .
7 . A pharmaceutical composition according to claim 1 , wherein said active ingredient is selected from the group consisting of:

8 . A pharmaceutical composition in unit dosage form comprising from 0.05 mg to 2000 mg of an active ingredient together with one or more pharmaceutically acceptable carriers or excipients, wherein said active ingredient is at least one compound according to formula (I):

10 . A pharmaceutical composition according to claim 8 wherein R 3 is —NR 8 R 9 , —C(═O)NR 8 R 9 or —OR 10 .
11 . A pharmaceutical composition according to claim 10 wherein R 3 is —NR 8 R 9 .

A pharmaceutical composition according to claim 8 wherein R 1 and R 2 together with the nitrogen to which they are attached, are forming a saturated or partially saturated bicyclic or tricyclic ring system containing from 4 to 12 carbon atoms and from 0 to 2 additional heteroatoms selected from nitrogen or oxygen, the ring system optionally being substituted with at least one of R 6 , hydroxyl or C 1 -C 6 alkyl, wherein the alkyl groups independently are optionally substituted with one ore more of R 7 .
13 . A pharmaceutical composition according to claim 8 wherein:
