Claims

Abstract

The present inventors have found that relaxin-3 has a feeding-stimulating activity, a body weight increasing activity, and fat weight increasing activity by intracerebroventricularly administering relaxin-3 to rats and observing the amount of feeding, body weight and fat weight, and so on after the administration. According to the present invention, a polypeptide having useful effects in stimulating feeding, increasing body weight, and fattening; a therapeutic agent containing said polypeptide; a method of screening for a compound, a substance, or a salt thereof which activates or suppresses a receptor of said polypeptide; a kit for said screening; and an agent which comprises a substance which inhibits expression of said polypeptide, such as a feeding-surpressing agent, a therapeutic agent for the treatment of obesity, and a therapeutic agent for the treatment of diabetes.

         53 . A method of screening for a compound which stimulates feeding or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell containing a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         54 . A method of screening for a compound which stimulates or suppresses feeding or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         55 . The method of screening for a compound which stimulates or suppresses feeding or a salt thereof according to  claim 54 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         56 . The method of screening according to any one of  claims 53  to  55 , wherein the relaxin-3 receptor is SALPR. 
     
     
         57 . The method of screening according to  claim 56 , wherein SALPR is a polypeptide containing the amino acid sequence represented by SEQ ID NO: 4. 
     
     
         58 . A kit for screening for a compound which stimulates feeding or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         59 . A kit for screening for a compound which stimulates or suppresses feeding or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         60 . The kit for screening for a compound which stimulates or suppresses feeding or a salt thereof according to  claim 59 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         61 . The kit for screening according to  claim 58 ,  59 , or  60 , wherein the relaxin-3 receptor is SALPR or its partial polypeptide. 
     
     
         62 . The kit for screening according to  claim 61 , wherein SALPR is a polypeptide comprising the amino acid sequence represented by SEQ ID NO: 4. 
     
     
         63 . A method of recovering feeding and/or body weight gain in a patient having a disease involving reduced feeding and/or weight loss, comprising administering an effective amount of relaxin-3, or a salt thereof, to a patient in need thereof. 
     
     
         64 . A method of screening for a compound which increases body weight or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell containing a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         65 . A method of screening for a compound which increases or decreases body weight or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         66 . The method of screening for a compound which increases or decreases body weight or a salt thereof according to  claim 65 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         67 . The method of screening according to any one of  claims 64  to  66 , wherein the relaxin-3 receptor is SALPR or it partial polypeptide. 
     
     
         68 . The method of screening according to  claim 67 , wherein SALPR is a polypeptide comprising the amino acid sequence represented by SEQ ID NO: 4. 
     
     
         69 . A kit for screening for a compound which increases body weight or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell containing a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         70 . A kit for screening for a compound which increases or decreases body weight or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         71 . The kit for screening for a compound which increases or decreases body weight or a salt thereof according to  claim 70 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         72 . The kit for screening according to  claim 69 ,  70  or  71 , wherein the relaxin-3 receptor is SALPR or its partial polypeptide. 
     
     
         73 . The kit for screening according to  claim 72 , wherein SALPR is a polypeptide comprising the amino acid sequence represented by SEQ ID NO: 4. 
     
     
         74 . A method of screening for a compound involved in the control of obesity or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell comprising a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         75 . A method of screening for a compound involved in the control of obesity or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         76 . The method of screening for a compound involved in the control of obesity or a salt thereof according to  claim 75 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         77 . The method of screening according to any one of  claims 74  to  76 , wherein the relaxin-3 receptor is SALPR or its partial polypeptide. 
     
     
         78 . The method of screening according to  claim 77 , wherein SALPR is a polypeptide comprising the amino acid sequence represented by SEQ ID NO: 4. 
     
     
         79 . A kit for screening for a compound involved in the control of obesity or a salt thereof, comprising the steps of
 (A) contacting a test substance with a relaxin-3 receptor, a cell containing a relaxin-3 receptor, or a membrane fraction of said cell, and   (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         80 . A kit for screening for a compound involved in the control of obesity or a salt thereof, comprising the step of
 (A) contacting relaxin-3, or a salt thereof, and a test substance with a relaxin-3 receptor, a cell which contains a relaxin-3 receptor, or a membrane fraction of said cell.   
     
     
         81 . The kit for screening for a compound involved in the control of obesity or a salt thereof according to  claim 80 , wherein it comprises the step of
 (B) measuring a cell-stimulating activity via the relaxin-3 receptor.   
     
     
         82 . The method of screening according to any one of  claims 79  to  81 , wherein the relaxin-3 receptor is SALPR or its partial polypeptide. 
     
     
         83 . The kit for screening according to  claim 82 , wherein SALPR is a polypeptide comprising the amino acid sequence represented by SEQ ID NO: 4.
