The present invention provides a 2-amino-5-heteroaryl-5-phenylimidazolone compound of formula I The present invention also provides methods for the use thereof to inhibit β-secretase (BACE) and treat β-amyloid deposits and neurofibrillary tangles

wherein W is CO, CS or CH 2 ;
X is N, NR, NO, S, SO m , O or CR 9 ;
Y is N, NR, NO, S, SO m , O or CR 10 ;
Z is C, N, NR, NO, S, SO m , O, CR 11 or CR 11 R 12 with the proviso that at least one of X, Y or Z must be N, NR, NO, S, SO m or O;
m is 1 or 2;
R is H or a C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 8 cycloalkyl or aryl(C 1 -C 6 )alkyl group each optionally substituted;
R 1 and R 2 are each independently H, COR 14 , CO 2 R 15 or an optionally substituted C 1 -C 4 alkyl group;

2 . The compound according to claim 1 wherein W is CO; X is CR 9 ; Y is CR 10 and Z is NR or S.
3 . The compound according to claim 1 wherein W is CO; X is N; Y is NR and Z is CR 11 R 12 .
4 . The compound according to claim 1 wherein R 7 is phenyl or heteroaryl.
5 . The compound according to claim 2 wherein R is H or C 1 -C 4 alkyl and R 4 is CN or COR 16 .
6 . The compound according to claim 2 wherein R 7 is phenyl or heteroaryl.
7 . The compound according to claim 3 wherein R 7 is phenyl or heteroaryl.
8 . The compound according to claim 6 wherein R 1 and R 2 are H; and R 3 is methyl.
9 . The compound according to claim 1 selected from the group consisting essentially of:

10 . A method for the treatment of a disease or disorder associated with excessive BACE activity in a patient in need thereof which comprises providing to said patient a therapeutically effective amount of a compound of formula I

11 . The method according to claim 10 wherein said disease or disorder is selected from the group consisting of: Alzheimer's disease; cognitive impairment; Down's Syndrome; HCHWA-D; cognitive decline; senile dementia; cerebral amyloid angiopathy; and a neurodegenerative disorder.
12 . The method according to claim 10 wherein said disease or disorder is characterized by the production of β-amyloid deposits or neurofibrillary tangles.
13 . A method for modulating the activity of BACE which comprises contacting a receptor thereof with an effective amount of a compound of claim 1 .
14 . A method for the treatment of Alzheimer's disease in a patient in need thereof which comprises providing to said patient an effective amount of a compound of claim 1 .
15 . A pharmaceutical composition which comprises a pharmaceutically acceptable carrier and an effective amount of a compound of formula I
