Abstract
The invention relates to variants of consensus interferon protein with improved properties, such as improved anti-viral activity, and use thereof. The variants are also easier to renature after denaturant treatment. The invention also relates to the preparation method of the variants consensus interferon.
Claims
exact text as granted — not AI-modified1 . A variant consensus interferon protein comprising the amino acid sequence of SEQ ID NO: 3.
2 . The variant consensus interferon protein of claim 1 , further comprising a modification at position S156, wherein said modification is S156C.
3 . The variant consensus interferon protein of claim 2 , further comprising at least one polyethylene glycol (PEG) moiety covalently attached to the protein.
4 . The variant consensus interferon protein of claim 3 , wherein said (PEG) moiety is attached at the S156C position of the protein.
5 . The variant consensus interferon protein of claim 3 or 4 , wherein said PEG moiety is (PEG)n.
6 . The variant consensus interferon protein of any one of claims 2 - 5 , wherein said PEG moiety is a PEG derivative.
7 . A DNA encoding the consensus interferon variant of claim 1 or 2 .
8 . An expression vector comprising the DNA of claim 7 .
9 . A host cell comprising the DNA of claim 7 .
10 . A host cell transformed with the expression vector of claim 8 .
11 . The host cell of claim 9 or 10 , wherein said host cell is a prokaryote cell.
12 . The host cell of claim 11 , wherein said host cell is E. coli.
13 . A pharmaceutical composition comprising the variant consensus interferon protein according to any one of claim 1 and a pharmaceutically acceptable excipient.
14 . A method of manufacturing a consensus interferon, the method comprising the steps of:
growing the host cell of claim 9 under a condition that the interferon consensus variants can be expressed; harvesting the host cell; and purifying the interferon consensus variants.
15 . A method of suppressing viral activity in a subject, the method comprising administering an animal or human subject with the variant consensus interferon protein according to any one of claim 1 .Cited by (0)
