Abstract

This document provides methods and materials relating to platelet lysates. For example, methods and materials for using platelet lysate compositions to grow adult stem cells, to differentiate adult stem cells, to grow primary cell cultures, to grow tumor stem cells, and to identify effective growth factors are provided.

         96 . A platelet lysate composition comprising a filtrate from a lysed platelet preparation passed through a 0.45 μm or smaller filter. 
     
     
         97 . The platelet lysate composition of  claim 96 , wherein said lysed platelet preparation is a lysed apheresis platelet preparation. 
     
     
         98 . The platelet lysate composition of  claim 96 , wherein said platelet lysate composition comprises greater than 200 pg of VEGF polypeptide per mL.

         112 . The isolated cell culture of  claim 111 , wherein said mesenchymal stem cells are adipose-derived, mesenchymal stem cells.

A method for obtaining a platelet plasma culture supplement comprising removing platelets or platelet debris from a platelet apheresis preparation such that at least some of the plasma within said platelet apheresis preparation is retained, thereby obtaining said platelet plasma culture supplement, wherein:
 (a) said platelet plasma culture supplement comprises the ability to induce proliferation of less than 1×10 6  adipose-derived, mesenchymal stem cells at passage five into at least 1×10 8  cells within 20 days when added to minimal essential medium at a concentration of 5 percent, or (b) minimal essential medium having 5 percent of said platelet plasma culture supplement is capable of establishing, from newly diagnosed human glioblastoma multiforme biopsy tissue, human glioma cultures having greater than 3×10 7  cells within 60 days at 37° C. with a success rate that is greater than 30 percent.   
     
     
         114 . The method of  claim 113 , wherein said platelet plasma culture supplement comprises the ability to induce proliferation of less than 1×10 6  adipose-derived, mesenchymal stem cells at passage five into at least 1×10 8  cells within 15 days when added to minimal essential medium at a concentration of 5 percent. 
     
     
         115 . The method of  claim 113 , wherein minimal essential medium having 5 percent of said platelet plasma culture supplement is capable of establishing, from newly diagnosed human glioblastoma multiforme biopsy tissue, human glioma cultures having greater than 3×10 7  cells within 60 days at 37° C. with a success rate that is greater than 30 percent, and wherein said success rate is greater than 40 percent.
