Claims

Abstract

This disclosure concerns novel compounds of Formula (I) or as defined in the specification and compositions comprising such novel compounds. These compounds are useful antiviral agents, especially in inhibiting the function of the NS5A protein encoded by Hepatitis C virus (HCV). Thus, the disclosure also concerns a method of treating HCV related diseases or conditions by use of these novel compounds or a composition comprising such novel compounds.

 wherein any said aryl and heteroaryl may optionally be substituted with one or more substituents independently selected from alkenyl, alkyl, haloalkyl, arylalkyl, heterocyclyl, heterocyclylalkyl, halogen, cyano, nitro, —C(O)OR 4 , OR 5 , —NR a R b , (NR a R b )alkyl, and (MeO)(HO)P(O)O—, and 
 wherein any said cycloalkyl and heterocyclyl may optionally be fused onto an aromatic ring and may optionally be substituted with one or more substituents independently selected from alkoxy, alkyl, hydroxyl, halogen, aryl, —NR a R b , oxo, and —C(O)OR 4 ; 
 R 3  is hydrogen, alkyl, or arylalkyl; 
 R 4  is selected from alkyl and arylalkyl; 
 R 5  is hydrogen, alkyl, or arylalkyl; 
 R a  and R b  are independently selected from hydrogen, alkyl, cycloalkyl, arylalkyl, heteroaryl, —C(O)R 6 , —C(O)OR 7 , —C(O)NR c R d , and (NR c R d )alkyl, or alternatively, R a  and R b , together with the nitrogen atom to which they are attached, form a five- or six-membered ring or bridged bicyclic ring structure, wherein said five- or six-membered ring or bridged bicyclic ring structure optionally may contain one or two additional heteroatoms independently selected from nitrogen, oxygen, and sulfur and may contain one, two, or three substituents independently selected from C 1  to C 6  alkyl, C 1  to C 4  haloalkyl, aryl, hydroxyl, C 1  to C 6  alkoxy, C 1  to C 4  haloalkoxy, and halogen; 
 R 6  is alkyl;

         9 . The compound of  claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 2  and R 22  are both hydrogen. 
     
     
         10 . A compound, or a pharmaceutically acceptable salt thereof, selected from the group consisting of:
 (2S,2′S)-tert-Butyl 2,2′-(4,4′-(2,2′-bithiophene-5,5′-diyl)bis(1H-imidazole-4,2-diyl))dipyrrolidine-1-carboxylate;   5′-(2,2′-Bithiene-5,5′-diyl)bis(2-((2S)-2-pyrrolidinyl)-1H-imidazole);   Di-tert-butyl (2S,2′S)-2,2′-(5,5′-bi-1,3-thiazole-2,2′-diylbis(1H-imidazole-5,2-diyl))di(1-pyrrolidinecarboxylate);   2,2′-Bis(2-((2S)-2-pyrrolidinyl)-1H-imidazol-5-yl)-5,5′-bi-1,3-thiazole;   tert-Butyl (2S)-2-(4-(5′-(2-((2S)-1-(tert-butoxycarbonyl)-2-pyrrolidinyl)-1H-imidazol-5-yl)-2,2′-bi-1,3-thiazol-5-yl)-1H-imidazol-2-yl)-1-pyrrolidinecarboxylate; and   5,5′-Bis(2-((2S)-2-pyrrolidinyl)-1H-imidazol-4-yl)-2,2′-bi-1,3-thiazole;   corresponding stereoisomers and tautomers thereof.

         12 . A composition comprising a compound of  claim 1 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier. 
     
     
         13 . A method of treating an HCV infection in a patient, comprising administering to the patient a therapeutically effective amount of a compound of  claim 1 , or a pharmaceutically acceptable salt thereof.
