Abstract

This disclosure provides a novel tumor suppressor, referred to as p28ING5, nucleic acid molecules encoding this protein, and methods of making and using these molecules. Also provided are methods of ameliorating, treating, detecting, prognosing, and diagnosing diseases and conditions associated with abnormal p28ING5 expression, such as neoplasia. Kits are also provided.

1 . A method of inhibiting cellular proliferation, comprising:
 contacting a cell with a p28ING5 recombinant nucleotide consisting of the nucleotide sequence of SEQ ID NO: 1 or a purified p28ING5 tumor suppressor protein having a sequence comprising the amino acid sequence set forth as SEQ ID NO: 2, thereby inhibiting cellular proliferation.   
     
     
         2 . The method of  claim 1 , wherein the purified p28ING5 tumor suppressor protein has a sequence consisting of the amino acid sequence set forth as SEQ ID NO: 2. 
     
     
         3 . The method of  claim 1 , wherein the cell is a tumor cell. 
     
     
         4 . The method of  claim 1 , wherein the method is an in vivo method of inhibiting cellular proliferation. 
     
     
         5 . The method of  claim 4 , wherein contacting a cell comprises administering to a cell in a subject an effective amount of a composition comprising the purified p28ING5 tumor suppressor protein having the amino acid sequence set forth as SEQ ID NO: 2 to inhibit cellular proliferation as compared to cellular proliferation in the absence of the composition, thereby inhibiting cellular proliferation in vivo. 
     
     
         6 . The method of  claim 5 , wherein the cell is a tumor cell and the method inhibits cellular proliferation of a tumor cell in vivo. 
     
     
         7 . The method of  claim 5 , wherein the purified p28ING5 tumor suppressor protein has a sequence consisting of the amino acid sequence set forth as SEQ ID NO: 2. 
     
     
         8 . The method of  claim 4 , wherein contacting a cell comprises administering to a cell in a subject an effective amount of a composition comprising the p28ING5 recombinant nucleotide consisting of the nucleotide sequence of SEQ ID NO: 1 to inhibit cellular proliferation as compared to cellular proliferation in the absence of the composition, thereby inhibiting cellular proliferation in vivo. 
     
     
         9 . The method of  claim 1 , wherein the p28ING5 recombinant nucleotide is operably linked to a promoter sequence. 
     
     
         10 . The method of  claim 1 , wherein contacting a cell comprises transfecting a cell with an expression vector comprising a promoter sequence operably linked to the p28ING5 recombinant nucleotide consisting of the nucleotide sequence of SEQ ID NO: 1. 
     
     
         11 . The method of  claim 10 , wherein the cell is a tumor cell.
