Abstract

A matrix, including epithelial basement membrane, for inducing repair of mammalian tissue defects and in vitro cell propagation derived from epithelial tissues of a warm-blooded vertebrate.

         18 . A method for manufacture of a tissue graft composition, comprising:
 providing a mammalian epithelial tissue comprising an epithelial basement membrane;   deepthelializing the mammalian epithelial tissue by removing epithelial cells and cellular elements of said epithelial cells from the epithelial basement membrane; and,   abrading said mammalian epithelial tissue on the surface opposite to said epithelial basement membrane to remove one or more abluminal layers of said mammalian epithelial tissue, thereby forming a tissue graft composition comprising the epithelial basement membrane free of said epithelial cells and said cellular elements of said epithelial cells, and one or more abluminal layers of said mammalian epithelial tissue.   
     
     
         19 . The method of  claim 18  wherein deepithelialization comprises placing said tissue in a deepithelializing solution comprising 1.0 N saline. 
     
     
         20 . The method of  claim 18  wherein said abrasive step comprises removal of tunica submucosa and tunica muscularis. 
     
     
         21 . The method of  claim 18  wherein deepithelialization is non-abrasive. 
     
     
         22 . The method of  claim 18  wherein said abrasive step comprises removal of the tunica muscularis. 
     
     
         23 . The method of  claim 18  wherein said mammalian epithelial tissue comprises urinary bladder. 
     
     
         24 . The method of  claim 18  wherein said mammalian epithelial tissue comprises small intestine. 
     
     
         25 . The method of  claim 18  wherein said mammalian epithelial tissue comprises intestine. 
     
     
         26 . The method of  claim 18  further comprising plating cells on the epithelial basement membrane free of epithelial cells and epithelial cellular elements. 
     
     
         27 . The method of  claim 26  wherein said cells comprise cells from a cultured cell line. 
     
     
         28 . The method of  claim 18  further comprising custom-shaping said tissue graft composition to conform to a diseased, damaged, or defective tissue. 
     
     
         29 . The method of  claim 18  further comprising solubilizing said tissue graft composition to form an injectable form of tissue graft composition. 
     
     
         30 . The method of  claim 18  further comprising combining said tissue graft composition with a pharmaceutical agent. 
     
     
         31 . The method of  claim 18  wherein said tissue graft composition is suturable.
