Abstract

As disclosed herein, the present invention provides for kits and a composition for diagnosis, prognosis, drug discovery, drug development, and patient stratification. The kits can comprise a plurality of binding elements for cell surface markers, and a plurality of binding elements for state-specific intracellular markers. The kits can further comprise a plurality of modulators directed for the particular cell function or signaling pathways. The kits can further include fixatives, permeabilizing agent, buffers, containers, instructions, and software for data analysis/compilation.

         36 . A kit comprising:
 (i) at least one binding element specific for a cell surface marker, wherein said cell surface marker is: CD11b, CD15, CD16, CD34, CD45, CD33, CD14, or CD7;   (ii) at least three binding elements specific for three of the following intracellular markers: cleaved PARP, p-CHK1, p-CHK2, p-H2AX, p-ATM, or p-ATR; and   (iii) at least two of the following modulators: Etoposide, Ara-C, Daunorubicin, Gemtuzumab, Apo2L/TRAIL, or FASL.   
     
     
         37 . A kit in accordance with  claim 36  further comprising:
 a binding element specific for p-p53, cleaved Caspase 8, or cleaved Caspase 3; and 
 at least one of the following modulators: Staurosporine or TNFα. 
 
     
     
         38 . A kit in accordance with  claim 36 , wherein the binding elements are antibodies. 
     
     
         39 . A kit in accordance with  claim 36 , further comprising a control cell and/or control compound. 
     
     
         40 . A kit in accordance with  claim 36 , further comprising a protocol or instructions. 
     
     
         41 . A kit in accordance with  claim 36 , further comprising software for data analysis. 
     
     
         42 . A kit in accordance with  claim 36 , further including a fixative, permeabilizing agent, buffer, instructions, detection element, and software for data analysis/compilation. 
     
     
         43 . A kit in accordance with  claim 42 , wherein the detection element comprises a fluorescent label or a label that is detectable by mass spectrometry. 
     
     
         44 . A method for classifying cells comprising:
 providing a kit in accordance with  claim 36 ;   contacting cells with two or more of the modulators of (iii) to activate cell signaling pathways;   contacting cells with at least one of the binding elements of (i) to identify a cell type;   contacting cells with three or more of the binding elements of (ii) to identify the one or more activated cell signaling pathways; and   classifying cells based on the identified cell type and the identified one or more activated cell signaling pathways.   
     
     
         45 . The method of  claim 44 , wherein the classification of cells is used in diagnosis or prognosis of a condition, or drug discovery, drug development or patient stratification for a condition. 
     
     
         46 . The method of  claim 45 , wherein said condition is one of non-Hodgkin's lymphoma, Hodgkin's lymphoma, non-B cell lymphomas, other lymphomas, acute or chronic leukemias, polycythemias, thrombocythemias, myelodysplastic disorders, myeloproliferative disorders, myelofibroses, atypical immune lymphoproliferations or a plasma cell disorder. 
     
     
         47 . The method of  claim 46 , wherein said plasma cell disorder is multiple myeloma, amyloidosis or Waldenstrom's macroglobulinemia. 
     
     
         48 . The method of  claim 46 , wherein said one of acute or chronic leukemias is a myeloid leukemia or a lymphoid leukemia. 
     
     
         49 . The method of  claim 48 , wherein said myeloid leukemia is acute myeloid leukemia (AML), chronic myelogenous leukemia (CML), or juvenile myelomonocytic leukemia (JMML). 
     
     
         50 . The method of  claim 48 , wherein said lymphoid leukemia is non-B cell acute lymphocytic leukemia (T-ALL), B cell acute lymphoblastic leukemia, or chronic lymphocytic leukemia (CLL). 
     
     
         51 . The method of  claim 45 , wherein said condition is a hematologic disease or disorder selected from the group consisting of myelodysplastic disorders, myeloproliferative disorders, myelofibroses, polycythemias, thrombocythemias, B cell immunoproliferations, post transplant lymphoproliferation disorder (PTLD), and non-B atypical immune lymphoproliferations. 
     
     
         52 . The method of  claim 45 , wherein said condition is CLL. 
     
     
         53 . The method of  claim 45 , wherein said condition is AML. 
     
     
         54 . The method of  claim 45 , wherein said condition is acute lymphocytic leukemia (ALL). 
     
     
         55 . The method of  claim 45 , wherein said condition is CML. 
     
     
         56 . The method of  claim 45 , wherein said condition is myelodysplastic syndrome (MDS).
