Abstract

Human monoclonal antibodies that specifically bind to rabies virus, antigen binding portions thereof, and methods of making and using such antibodies and antigen binding portions thereof for treating rabies virus in a subject, are provided herein.

1 . An isolated monoclonal antibody that competes for binding to rabies virus G protein with an antibody comprising:
 (a) a heavy chain variable region CDR3 sequence comprising SEQ ID NO:5;   (b) a light chain variable region CDR3 sequence comprising SEQ ID NO:8;   (c) a heavy chain variable region CDR2 sequence comprising SEQ ID NO:4;   (d) a light chain variable region CDR2 sequence comprising SEQ ID NO:7;   (e) a heavy chain variable region CDR1 sequence comprising SEQ ID NO:3; and   (f) a light chain variable region CDR1 sequence comprising SEQ ID NO:6.   
     
     
         2 . An isolated monoclonal antibody that binds to binds to the same or an overlapping epitope on rabies virus G protein as an antibody comprising:
 (a) a heavy chain variable region CDR3 sequence comprising SEQ ID NO:5;   (b) a light chain variable region CDR3 sequence comprising SEQ ID NO:8;   (c) a heavy chain variable region CDR2 sequence comprising SEQ ID NO:4;   (d) a light chain variable region CDR2 sequence comprising SEQ ID NO:7;   (e) a heavy chain variable region CDR1 sequence comprising SEQ ID NO:3; and   (f) a light chain variable region CDR1 sequence comprising SEQ ID NO:6.   
     
     
         3 . The isolated monoclonal antibody of  claim 1 , wherein the antibody comprises a variable heavy chain region comprising SEQ ID NO:1 and a variable light chain region comprising SEQ ID NO:2. 
     
     
         4 . The isolated monoclonal antibody of  claim 1 , wherein the antibody comprises a human, humanized or chimeric antibody. 
     
     
         5 . The isolated monoclonal antibody of  claim 1 , wherein the antibody comprises a full length antibody. 
     
     
         6 . The isolated monoclonal antibody of any  claim 1 , wherein the antibody is selected from the group consisting of a Fab, F(ab′)2, Fv or a single chain Fv fragment. 
     
     
         7 . A composition comprising the antibody of  claim 1  in a pharmaceutically acceptable carrier. 
     
     
         8 . The composition of  claim 7 , further comprising one or more additional antibodies. 
     
     
         9 . A method of treating rabies virus disease in a subject, the method comprising: administering to the subject the antibody of  claim 1  in an amount effective to inhibit a symptom of rabies virus disease. 
     
     
         10 . The method of  claim 9 , wherein the antibody is administered in combination with one or more additional antibodies. 
     
     
         11 . A method of producing the antibody of  claim 1 , comprising transfecting a host cell with one or more vectors encoding the heavy and light light chains of the antibody and isolating the expressed antibody.
