Abstract
Disclosed are therapeutic, blood-derived activated leukocyte compositions, methods of making them, and methods of using the compositions to repair or promote the prevention and healing of wounds.

21 . A composition comprising activated leukocytes, wherein the composition comprises:
a) about 40% to about 90% granulocytes; b) about 5% to about 20% monocytes; and c) about 5% to about 30% lymphocytes.
22 . The composition of claim 21 , wherein the granulocytes comprise:
a) about 52% to about 78% neutrophils; b) about 1% to about 9% eosinophils; and c) about 1% to about 2% basophils.
23 . The composition of claim 21 , wherein the lymphocytes comprise:
a) about 7% to about 25% B cells (CD19+); b) about 20% to about 30% NK cells (CD3−/CD56+); c) about 40% to about 60 T cells (CD3+); d) about 0% to about 30 of NKT cells (CD3+/CD56+); e) about 8% to about 20% of T helper cells, (CD4+/CD3+); and f) about 20% to about 30% of CD8+/CD3+ cells.
24 . The composition of claim 21 , wherein the composition includes T-helper cells and T-suppressor cells in a ratio of less than 0.8.
25 . The composition of claim 21 , further comprising mesenchymal stem cells in an amount ranging from about 0.1% to about 5.0% of the total cell population in the composition.
26 . The composition of claim 21 , further comprising endothelial progenitor cells in a amount ranging from about 0.1% to about 5.0% of the total cell population in the composition.
27 . The composition of claim 21 , further comprising platelets.
28 . An article of manufacture comprising:
a composition comprising activated leukocytes, wherein the composition comprises:
a) about 40% to about 90% granulocytes;
b) about 5% to about 20% monocytes; and
c) about 5% to about 30% lymphocytes; and
a dressing.
29 . The article of manufacture of claim 28 , wherein the dressing is a dry dressing, a moisture-keeping barrier dressing, or a bioactive dressing.
30 . The article of manufacture of claim 29 , wherein the dry dressing is gauze, a bandage, a non-adhesive mesh, a membrane, foils, foam, or a tissue adhesive.
31 . The article of manufacture of claim 29 , wherein the moisture-keeping barrier dressing is a paste, a cream, an ointment, a nonpermeable or semi-permeable membrane or foil, a hydrocolloid, a hydrogel, or combinations thereof.
32 . The article of manufacture of claim 29 , wherein the bioactive dressing is an antimicrobial dressing.
33 . The composition of claim 21 , further comprising as a matrix or scaffold a material suitable for implantation in a person.
34 . The composition of claim 33 , wherein the material is a solid before implantation.
35 . The composition of claim 33 , wherein the material is a gel that solidifies following implantation.
36 . A method for treating a wound comprising administering to the wound a composition comprising activated leukocytes, wherein the composition comprises:
a) about 40% to about 90% granulocytes; b) about 5% to about 20% monocytes; and c) about 5% to about 30% lymphocytes.

The method of claim 36 , wherein the wound is a decubital ulcer, a pressure ulcer, a lower extremity ulcer, a deep sternal wound, a post-operative wound, a refractory post-operative wound of the trunk area, a wound to the great saphenous vein following harvesting of the great saphenous vein, a venous ulcer, or an anal fissure.
38 . The method of claim 36 , wherein wound is a lower extremity ulcer in a diabetic patient.
39 . The method of claim 36 , wherein the wound is a venous ulcer, pressure ulcer, or post-operative ulcer.
40 . A method of inhibiting the onset of infection in a wound, comprising administering to the wound a composition comprising activated leukocytes, wherein the composition comprises:
a) about 40% to about 90% granulocytes; b) about 5% to about 20% monocytes; and c) about 5% to about 30% lymphocytes.
41 . The method of claim 40 , wherein the wound is caused by trauma.
42 . The method of claim 41 , wherein the trauma is caused by surgery.
