Abstract
The present invention provides compounds and methods for treating or preventing pulmonary diseases include COPD and asthma. In particular, the present invention provides for compounds comprising type V collagen, or tolerizing fragments thereof, for the treatment of COPD and asthma.

1 . A composition for treating chronic obstructive pulmonary disease in a COPD patient comprising a therapeutically effective amount of type V collagen, or a tolerogenic fragment thereof.
2 . The composition of claim 1 wherein the COPD patient has emphysema or chronic obstructive bronchitis.
3 . The composition of claim 1 wherein the type V collagen or tolerogenic fragment thereof is prepared for oral administration.
4 . The composition of claim 3 comprising between 0.1 mg and 0.5 mg of type V collagen.
5 . The composition of claim 1 wherein the type V collagen or tolerogenic fragment thereof is prepared for intravenous administration, intrapulmonary instillation, administration by inhalation, or for intramuscular administration.
6 . The composition of claim 1 , further comprising a bronchodilator or for use in conjunction with a bronchodilator.
7 . The composition of claim 1 further comprising a corticosteroid or for use in conjunction with a corticosteroid.
8 . A composition for treating asthma comprising a therapeutically effective amount of type V collagen, or a tolerogenic fragment thereof.
9 . The composition of claim 8 wherein the type V collagen or tolerogenic fragment thereof is prepared for intravenous administration, intrapulmonary instillation, administration by inhalation, or for intramuscular administration.
10 . The composition of claim 9 wherein the type V collagen is administered at a dose of between 0.1 mg and 0.5 mg.
11 . The composition of claim 8 further comprising a corticosteroid or for use in conjunction with a corticosteroid.
12 . The composition of claim 8 further comprising a bronchodilator or for use in conjunction with a bronchodilator.
13 . The composition of claim 8 further comprising a leukotriene modifier or for use in conjunction with a leukotriene modifier.
14 . A composition for preventing the development of chronic obstructive pulmonary disease in a subject at risk for developing chronic obstructive pulmonary disease comprising a therapeutically effective amount of type V collagen, or a tolerogenic fragment thereof.
15 . The composition of claim 14 wherein the type V collagen or tolerogenic fragment thereof is administered orally.
16 . The composition of claim 15 wherein the type V collagen is administered at a dose of between 0.1 mg and 0.5 mg.
17 . The composition of claim 14 wherein the type V collagen or tolerogenic fragment thereof is prepared for intravenous administration, intrapulmonary instillation, administration by inhalation, or for intramuscular administration.
18 . A composition for preventing the development or worsening of asthma in a subject at risk for developing asthma comprising a therapeutically effective amount of type V collagen, or a tolerogenic fragment thereof.
19 . The composition of claim 18 wherein the type V collagen or tolerogenic fragment thereof is administered orally.
20 . The composition of claim 19 wherein the type V collagen is administered at a dose of between 0.1 mg and 0.5 mg.
21 . The composition of claim 18 wherein the type V collagen or tolerogenic fragment thereof is prepared for intravenous administration, intrapulmonary instillation, administration by inhalation, or for intramuscular administration.
22 . A method for identifying a COPD or asthma patient as a candidate for collagen V tolerance therapy comprising,
contacting at least a portion of a sample of blood from the patient with collagen V or an antigenic fragment thereof; and measuring the level of antibodies that bind to the collagen V or antigenic fragment thereof; wherein the presence of antibodies specifically bound to the collagen V is indicative that the COPD or asthma patient would benefit from collagen V tolerance therapy.
23 . The method of claim 22 wherein the collagen V or antigenic fragment thereof is conjugated to a bead.
24 . The method of claim 22 wherein the measuring comprises
contacting the antibodies that specifically bind to the collagen V or antigenic fragment thereof with a fluorescently labeled anti-IgG antibody; and
detecting by flow cytometry the amount of fluorescently labeled anti-IgG antibody bound to the antibodies bound to the collagen V.
25 . A method for identifying an individual at risk for developing COPD or asthma, comprising:
contacting at least a portion of a sample of blood from the individual with collagen V or an antigenic fragment thereof; and measuring the level of antibodies that bind to the collagen V or an antigenic fragment thereof; wherein the presence of antibodies that specifically bind to the collagen V is associated with a higher risk than would be expected in an individual with no antibodies that bind to the collagen V.
26 . The method of claim 25 wherein the collagen V or antigenic fragment thereof is conjugated to a bead.
27 . The method of claim 25 wherein the measuring comprises
contacting the antibodies that bind to the collagen V or antigenic fragment thereof with a fluorescently labeled anti-IgG antibody; and
detecting by flow cytometry the amount of fluorescently labeled anti-IgG antibody bound to the antibodies bound to the collagen V.
28 . The method of claim 24 or 27 wherein the anti-IgG antibody detects all IgG subtypes.
29 . The method of claim 24 or 27 wherein the anti-IgG antibody specifically detects the IgG1 subtype.
30 . The method of claim 24 or 27 wherein the anti-IgG antibody specifically detects the IgG2 subtype.
31 . The method of claim 24 or 27 wherein the anti-IgG antibody specifically detects the IgG3 subtype.
32 . The method of claim 24 or 27 wherein the anti-IgG antibody specifically detects the IgG4 subtype.
33 . A method for monitoring the progression of COPD in an individual comprising,
contacting at least a portion of a first sample of blood from the individual with collagen V or an antigenic fragment thereof; measuring the level of antibodies that specifically bind to the collagen V or antigenic fragment thereof in the first sample of blood; contacting at least a portion of a second sample of blood from the individual taken at a later time point, with collagen V or an antigenic fragment thereof; measuring the level of antibodies that specifically bind to the collagen V or antigenic fragment thereof in the second sample of blood; and comparing the level of antibodies that specifically bind to the collagen V or antigenic fragment thereof in the second sample of blood to the level of antibodies that specifically bind to the collagen V or antigenic fragment thereof in the first sample of blood; wherein an increase in the level of antibodies bound to the collagen V in the second sample as compared to the first sample is indicative of worsening of COPD and a decrease in the level of antibodies bound to the collagen V in the second sample as compared to the first sample is indicative of amelioration of COPD.
34 . The method of claim 33 wherein the collagen V or antigenic fragment thereof is conjugated to a bead.
35 . The method of claim 33 wherein the measuring comprises
contacting the antibodies that bind to the collagen V or antigenic fragment thereof with a fluorescently labeled anti-IgG antibody; and
detecting by flow cytometry the amount of fluorescently labeled anti-IgG antibody bound to the antibodies bound to the collagen V.
36 . A method for monitoring the progression of asthma in an individual comprising,
contacting at least a portion of a first sample of blood from the individual with collagen V or an antigenic fragment thereof; measuring the level of antibodies that bind to the collagen V or antigenic fragment thereof in the first sample of blood; contacting at least a portion of a second sample of blood from the individual taken at a later time point, with collagen V or an antigenic fragment thereof; measuring the level of antibodies that bind to the collagen V or antigenic fragment thereof in the second sample of blood; and comparing the level of antibodies that bind to the collagen V or antigenic fragment thereof in the second sample of blood to the level of antibodies that bind to the collagen V or antigenic fragment thereof in the first sample of blood; wherein an increase in the level of antibodies bound to the collagen V in the second sample as compared to the first sample is indicative of worsening of asthma and a decrease in the level of antibodies bound to the collagen V in the second sample as compared to the first sample is indicative of amelioration of asthma.
37 . The method of claim 36 wherein the collagen V or antigenic fragment thereof is conjugated to a bead.
38 . The method of claim 37 wherein the measuring comprises
contacting the antibodies that bind to the collagen V or antigenic fragment thereof with a fluorescently labeled anti-IgG antibody; and
detecting by flow cytometry the amount of fluorescently labeled anti-IgG antibody bound to the antibodies bound to the collagen V.
39 . The method of claim 35 or 38 wherein the anti-IgG antibody detects all IgG subtypes.
40 . The method of claim 35 or 38 wherein the anti-IgG antibody specifically detects the IgG1 subtype.
41 . The method of claim 35 or 38 wherein the anti-IgG antibody specifically detects the IgG2 subtype.
42 . The method of claim 35 or 38 wherein the anti-IgG antibody specifically detects the IgG3 subtype.
