Abstract
Provided are compositions and methods for delivering biological moieties such as modified nucleic acids into cells to modulate protein expression. Such compositions and methods include the use of modified messenger RNAs, and are useful for production of proteins.
Claims
exact text as granted — not AI-modified1 . A method of producing an antibody in a human cell, comprising contacting said human cell with a composition comprising:
a. a first mRNA encoding the heavy chain of said antibody, and b. a second mRNA encoding the light chain of said antibody; wherein each of said first and said second mRNA contains at least one nucleoside modification and are each substantially devoid of uracil nucleotides.
2 . The method of claim 1 , wherein said at least one nucleoside modification is selected from the group consisting of pseudouridine.
3 . The method of claim 1 , wherein each of said first and second mRNA further contains the nucleoside modification 5-methyl-cytidine.
4 . The method of claim 1 , wherein the 5′ cap structure of each of said first and second mRNA is independently selected from the group consisting of Cap 1.
5 . The method of claim 1 , wherein the amount of antibody produced by said human cell is greater than the amount produced by a composition comprising said first and said second mRNA which are unmodified.
6 . The method of claim 1 , wherein in the composition the first mRNA and the second mRNA are in a 1:1 ratio.Cited by (0)
