Claims

Abstract

The invention provides a polypeptide containing at least one IgG Fc region, wherein said at least one IgG Fc region is glycosylated with at least one galactose moiety connected to a respective terminal sialic acid moiety by a α2,6 linkage, and wherein said polypeptide having a higher anti-inflammatory activity as compared to an unpurified antibody.

         32 . A method of inhibiting inflammation in a subject, comprising:
 providing a preparation comprising a modified IVIG composition prepared from an unmodified IVIG composition, wherein the modified IVIG composition has (i) increased anti-inflammatory activity as compared to the unmodified IVIG composition, and (ii) a higher content of α2,6 linked sialic acid in the N-linked glycans of Fc regions than the unmodified IVIG composition; and   administering to a subject having inflammation a dose of the preparation sufficient to inhibit inflammation in the subject.   
     
     
         33 . The method of  claim 32  wherein the modified IVIG composition was prepared by a method comprising treating the unmodified IVIG composition with α2-6 sialyltransferase. 
     
     
         34 . The method of  claim 32 , wherein the modified IVIG composition was prepared from the unmodified IVIG composition by method comprising the use of one or more of affinity chromatography HPLC, lectin affinity chromatography, high pH anion exchange chromatography, ion-exchange chromatography, and any combination thereof 
     
     
         35 . The method of  claim 32 , wherein the modified IVIG composition was prepared from the unmodified IVIG composition by a method comprising the use of a lectin having a higher affinity to α2,6 linkages than to α2,3 linkages between sialic acids and galactose. 
     
     
         36 . The method of  claim 32 , wherein the subject has an inflammatory disease. 
     
     
         37 . The method of  claim 32 , wherein the subject has arthritis, thrombocytopenia, or nephritis.
