Abstract
The subject invention provides efficient and convenient methods of isolating 9-oxo-10E, 12E-octadecadienoic acid methyl ester (9-KODE methyl ester) from C. versicolor. In addition, the subject invention provides therapeutic uses of Coriolus versicolor extracts, biologically-active chemical substituents isolated from C. versicolor, as well as 9-KODE methyl ester and related compounds. In a preferred embodiment, the subject invention can be used to inhibit the metastatic spread of cancer cells.

1 . A method of obtaining isolated 9-oxo-10E, 12E-octadecadienoic acid methyl ester (9-KODE methyl ester), comprising the steps of:
a) providing a sufficient quantity of raw material of Coriolus versicolor; and b) extracting the raw material of Coriolus versicolor with a polar solvent that is alcohol or an alcohol-water mixture at a temperature of about 15° C. to about 30° C. to yield a Coriolus versicolor alcohol extract and a residue, and recovering the Coriolus versicolor alcohol extract; and c) isolating 9-oxo-10E, 12E-octadecadienoic acid methyl ester from the Coriolus versicolor alcohol extract wherein step b) is performed once or more than once.
2 . The method, according to claim 1 , wherein the polar solvent is ethanol or a water-ethanol mixture.
3 . A compound having the chemical structure represented by formula I, or a salt thereof,
wherein is a double bond or a single bond;
R 1 is H, OH, a straight or branched chain C 1 to C 4 alkyl group, or OR a wherein R a is a straight or branched chain C 1 to C 4 alkyl group; and
R 2 is H, O, or a straight or branched chain C 1 to C 4 alkyl group; and
R 3 is H, OH, O, halo, a straight or branched chain C 1 to C 4 alkyl group, or OR a wherein R a is a straight or branched chain C 1 to C 4 alkyl group.
4 . The compound, according to claim 3 , wherein R 1 is OH, OCH 3 , or OC 2 H 5 .
5 . The compound, according to claim 3 , wherein R 2 is O.
6 . The compound, according to claim 3 , wherein R 3 is O or OH.
7 . The compound, according to claim 3 , which is 9-oxo-10E, 12E-octadecadienoic acid methyl ester.
8 . A composition comprising a compound of claim 3 .
9 . The composition, according to claim 8 , further comprising a therapeutically-acceptable carrier.
10 . The composition, according to claim 8 , comprising 9-oxo-10E, 12E-octadecadienoic acid methyl ester.
11 . A method for treating cancer or a tumor, comprising administering, to a subject in need of such treatment, an effective amount of a composition comprising a compound of formula I, or a salt thereof,
wherein is a double bond or a single bond;
R 1 is H, OH, a straight or branched chain C 1 to C 4 alkyl group, or OR a wherein R a is a straight or branched chain C 1 to C 4 alkyl group; and
R 2 is H, O, or a straight or branched chain C 1 to C 4 alkyl group; and
R 3 is H, OH, O, halo, a straight or branched chain C 1 to C 4 alkyl group, or OR a wherein R a is a straight or branched chain C 1 to C 4 alkyl group.
12 . The method of claim 11 , wherein the compound is 9-oxo-10E, 12E-octadecadienoic acid methyl ester.
13 . The method of claim 11 , wherein the subject is a human.
14 . The method of claim 11 , wherein the subject has metastatic cancer and the administration of the composition is used to reduce cancer metastasis.
15 . The method of claim 11 , wherein the cancer or tumor is selected from the group consisting of brain cancer or tumor, nasopharyngeal carcinoma, breast cancer or tumor, lung cancer or tumor, leukemia, lymphoma, colon cancer or tumor, liver cancer or tumor, stomach cancer or tumor, esophageal cancer or tumor, bladder cancer or tumor, and gastric cancer or tumor.
16 . The method of claim 11 , wherein the subject has brain cancer or tumor, and wherein the administration of the composition is used to treat brain cancer or tumor.
17 . The method of claim 11 , wherein the subject has breast cancer or tumor, and wherein the administration of the composition is used to treat breast cancer or tumor.
18 . The method of claim 11 , wherein the subject has lung cancer or tumor, and wherein the administration of the composition is used to treat lung cancer or tumor.
