Abstract

The present invention provides compositions and methods relating to or derived from anti-PAR-2 antibodies. In particular embodiments, the invention provides human antibodies that bind PAR-2, PAR-2-binding fragments and derivatives of such antibodies, and PAR-2-binding polypeptides comprising such fragments. Other embodiments provide nucleic acids encoding such antibodies, antibody fragments and derivatives and polypeptides, cells comprising such polynucleotides, methods of making such antibodies, antibody fragments and derivatives and polypeptides, and methods of using such antibodies, antibody fragments and derivatives and polypeptides, including methods of treating or diagnosing subjects having PAR-2-related disorders or conditions.

An isolated antigen binding protein having a heavy chain and a light chain, the heavy chain comprising three complementarity determining regions (CDRs) designated CDR1, CDR2 and CDR3, and the light chain comprising three complementarity determining regions designated CDR1, CDR2 and CDR3, wherein
 a) the heavy chain CDR1 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 31 through 35 of SEQ ID NO:9, amino acids 31 through 35 of SEQ ID NO:13 and amino acids 31 through 35 of SEQ ID NO:17;   b) the heavy chain CDR2 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 50 through 66 of SEQ ID NO:9, amino acids 50 through 66 of SEQ ID NO:13 and amino acids 50 through 66 of SEQ ID NO:17; and   c) the heavy chain CDR3 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 99 through 115 of SEQ ID NO:9, amino acids 99 through 115 of SEQ ID NO:13 and amino acids 99 through 115 of SEQ ID NO: 17; and      a′) the light chain CDR1 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 23 through 33 of SEQ ID NO:11, amino acids 23 through 33 of SEQ ID NO:15 and amino acids 23 through 33 of SEQ ID NO:19   b′) the light chain CDR2 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 49 through 55 of SEQ ID NO:11, amino acids 49 through 55 of SEQ ID NO: IS and amino acids 49 through 55 of SEQ ID NO: 19; and   c′) the light chain CDR3 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 88 through 97 of SEQ ID NO: 11, amino acids 88 through 97 of SEQ ID NO:15 and amino acids 88 through 97 of SEQ ID NO: 19.   
     
     
         6 . The antigen binding protein of  claim 5 , the heavy chain further comprising from one to four framework regions (FRs) designated FR1, FR2 FR3 and FR4, and the light chain further comprising from one to four framework regions designated FR1, FR2 FR3 and FR4, wherein
 a) the heavy chain FR1 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 1 through 30 of SEQ ID NO:9, amino acids I through 30 of SEQ ID NO: 13 and amino acids 1 through 25 of SEQ ID NO:17   b) the heavy chain FR2 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 36 through 49 of SEQ ID NO:9, amino acids 36 through 49 of SEQ ID NO: 13 and amino acids 36 through 49 of SEQ ID NO: 17;   c) the heavy chain FR3 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 67 through 98 of SEQ ID NO:9, amino acids 67 through 98 of SEQ ID NO:13 and amino acids 67 through 98 of SEQ ID NO: 17; and   d) the heavy chain FR4 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 116 though 126 of SEQ ID NO:9, amino acids 116 though 126 of SEQ ID NO:13 and amino acids 116 though 126 of SEQ ID NO:17 and   a′) the light chain FR1 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids I through 22 of SEQ ID NO:11, amino acids 1 through 22 of SEQ ID NO:15 and amino acids 1 through 22 of SEQ ID NO:19;   b′) the light chain FR2 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 34 through 48 of SEQ ID NO:11, amino acids 34 through 48 of SEQ ID NO:15 and amino acids 34 through 48 of SEQ ID NO:19;   c′) the light chain FR3 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 56 through 87 of SEQ ID NO:11, amino acids 56 through 87 of SEQ ID NO:15 and amino acids 56 through 87 of SEQ ID NO:19; and   d′) the light chain FR4 has an amino acid sequence selected from the group of amino acid sequences consisting of amino acids 98 through 107 of SEQ NO:11, amino acids 98 through 107 of SEQ ID NO:15 and amino acids 98 through 107 of SEQ ID NO:19.
