Claims

Abstract

Provided is a peptide containing a variable region and improved in production efficiency. The peptide contains a variable region to which an antigen-binding site is to be formed and has an amino acid sequence expressing a specific adsorption function to a solid phase at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region.

What is claimed is as follows: 
     
         1 . A method for producing a reconstructed peptide, characterized by bringing a solution, which contains (a) a denatured peptide having an amino acid sequence expressing a specific adsorption function to a solid phase, (b) a denaturizing agent, and (c) a contaminant, into contact with a surface of the solid-phase, thereby permitting the peptide to directly adsorb to the surface of the solid phase to reconstruct a tertiary structure of the denatured peptide. 
     
     
         2 . The method for producing a peptide according to  claim 1 , characterized in that the denatured peptide is denatured from a state of an inclusion body. 
     
     
         3 . The method for producing a peptide according to  claim 1 , characterized in that the peptide contains a variable region in which an antigen-binding site is to be formed and has the amino acid sequence at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region. 
     
     
         4 . The method for producing a peptide according to  claim 1 , characterized in that the solution contains a crushed product, a medium or a secretion of a host which produced the peptide. 
     
     
         5 . The method for producing a peptide according to  claim 1 , characterized in that the amino acid sequence is any one of amino acid sequences represented by SEQ ID NOs: 1 to 20 of the sequence listing. 
     
     
         6 . The method for producing a peptide according to  claim 1 , characterized in that the peptide is reconstructed a tertiary structure by the denaturing agent is removed while the peptide is directly adsorbed to the surface of the solid phase. 
     
     
         7 . The method for producing a peptide according to  claim 1 , characterized in that the amino acid sequence expresses a specific adsorption function to a solid phase of a resin surface hydrophilically treated, the peptide is directly adsorbed to the resin surface. 
     
     
         8 . The method for producing a peptide according to  claim 7 , characterized in that the resin surface is a hydrophilic polystyrene surface. 
     
     
         9 . A method for producing a solid phase having a peptide immobilized thereto, characterized by introducing a gene having a base sequence, which encodes a peptide having an amino acid sequence expressing a specific adsorption function to the solid phase, into a host to form a transformant, bringing a solution containing a product thereof into contact with a surface of the solid-phase to allow the peptide to directly adsorb to the surface of the solid-phase. 
     
     
         10 . The method for producing a solid phase having a peptide immobilized according to  claim 9 , characterized in that the peptide contains a variable region in which an antigen-binding site is to be formed and has the amino acid sequence at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region. 
     
     
         11 . The method for producing a solid phase having a peptide immobilized according to  claim 9 , characterized in that the amino acid sequence is any one of amino acid sequences represented by SEQ ID NOs: 1 to 20 of the sequence listing.

A method for producing a solid phase having a peptide immobilized thereto, characterized by bringing a solution, which contains (a) a denatured peptide having an amino acid sequence expressing a specific adsorption function to a solid phase, (b) a denaturizing agent, and (c) a contaminant, into contact with a surface of the solid-phase, thereby permitting the peptide to directly adsorb to the surface of the solid phase to reconstruct a tertiary structure of the denatured peptide.

The method for producing a solid phase having a peptide immobilized according to  claim 12 , characterized in that the peptide contains a variable region in which an antigen-binding site is to be formed and has the amino acid sequence at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region. 
     
     
         14 . The method for producing a solid phase having a peptide immobilized according to  claim 12 , characterized in that the amino acid sequence is any one of amino acid sequences represented by SEQ ID NOs: 1 to 20 of the sequence listing. 
     
     
         15 . The method for producing a solid phase having a peptide immobilized thereto according to  claim 12 , characterized in that the solution contains a crushed product, a medium or a secretion of a host which produced the peptide.

The method for producing a solid phase having a peptide immobilized according to  claim 15 , characterized in that the peptide contains a variable region in which an antigen-binding site is to be formed and has the amino acid sequence at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region. 
     
     
         17 . The method for producing a solid phase having a peptide immobilized according to  claim 15 , characterized in that the amino acid sequence is any one of amino acid sequences represented by SEQ ID NOs: 1 to 20 of the sequence listing. 
     
     
         18 . The method for producing a solid phase having a peptide immobilized thereto according to  claim 9 , characterized in that the host is yeast and the solution containing the product contains a secretion produced by secretion from the yeast. 
     
     
         19 . The method for producing a solid phase having a peptide immobilized according to  claim 18 , characterized in that the peptide contains a variable region in which an antigen-binding site is to be formed and has the amino acid sequence at a site closer to the C-terminal than a heavy-chain variable region or at a site closer to the C-terminal than a light-chain variable region. 
     
     
         20 . The method for producing a solid phase having a peptide immobilized according to  claim 18 , characterized in that the amino acid sequence is any one of amino acid sequences represented by SEQ ID NOs: 1 to 20 of the sequence listing.
