Claims

Abstract

Disclosed are compounds of Formula (I) and pharmaceutically acceptable salts and solvates thereof. Such compounds are MEK inhibitors and are useful in the treatment of proliferative diseases, such as cancer. Also disclosed are pharmaceutical compositions containing such compounds as well as methods of using the compounds and compositions of the invention in the treatment of cancer.

         45 . The method of  claim 44 , wherein X 1  is OH, and the reaction occurs in the presence of a coupling reagent. 
     
     
         46 . The method of  claim 45 , wherein the reaction occurs in the presence of (benzotriazol-1-yl-oxytripyrrolidinophosphonium hexafluorophosphate (PyBOP), N,N,N′,N′-tetramethyl-O-(1H-benzotriazol-1-yl)uranium hexafluorophosphate (HBTU) or 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDCI). 
     
     
         47 . The method of  claim 46 , wherein the reaction occurs in the presence of a base. 
     
     
         48 . The method of  claim 47 , wherein the reaction occurs in the presence of diisopropylethylamine (DIPEA), N-methylmorpholine, dimethylaminopyridine (DMAP) or triethylamine. 
     
     
         49 . The method of  claim 44 , wherein X 1  is halogen, and the reaction occurs in the presence of a base. 
     
     
         50 . The method of  claim 49 , wherein the reaction occurs in the presence of diisopropylethylamine (DIPEA) or sodium bicarbonate. 
     
     
         51 . The method of  claim 44 , wherein the reaction occurs in the presence of dimethyl formamide, tetrahydrofuran, 1,4-dioxane or dichloromethane. 
     
     
         52 . The method of  claim 44 , wherein A is phenylene optionally substituted with one or two groups selected from R 10 , R 12 , R 14 , and R 16  where R 10 , R 12 , R 14  and R 16  are independently hydrogen or halo;
