Abstract

The present invention provides compositions (i.e., preservation solutions) for preserving biologics comprising at least one membrane penetrable sugar, at least one membrane impenetrable sugar, at least one anti-microbial agent, at least one anti-oxidant, adenosine albutinum, a salt, a buffer, and a chelating agent, and to methods of using such preservation solutions to preserve biologies.

         18 . A method of preserving a cancer cell comprising:
 contacting the cancer cell with a solution, wherein the solution comprises:
 a buffer; 
 from about 0.01% to about 10% salt; 
 at least one membrane penetrable sugar; 
 a neutral dextran with a molecular weight greater than or equal to 50,000 daltons; and 
 0.1% w/v to about 12% w/v glucose; and 
   drying the cancer cell by room temperature induced dryness.   
     
     
         19 . The method of  claim 18 , wherein the buffer is from about 0.1 mg/mL, to about 2.0 mg/mL, phosphate buffer. 
     
     
         20 . The method of  claim 18 , wherein the buffer is 0.1 M HEPES. 
     
     
         21 . The method of  claim 18 , wherein the salt is from about 0.25% to about 2% NaCl. 
     
     
         22 . The method of  claim 18 , wherein the at least one membrane penetrable sugar is trehalose. 
     
     
         23 . The method of  claim 22 , wherein trehalose is 0.1% w/v to about 12% w/v. 
     
     
         24 . The method of  claim 18 , wherein the neutral dextran with a molecular weight greater than or equal to 50,000 daltons is Dextran-70. 
     
     
         25 . The method of  claim 24 , wherein the Dextran-70 is from about 0.01% w/v to about 25% w/v. 
     
     
         26 . The method of  claim 24 , wherein the Dextran-70 is 6% w/v. 
     
     
         27 . The method of  claim 18 , wherein the glucose is about 2% w/v. 
     
     
         28 . The method of  claim 18 , further comprising rehydrating the cancer cell. 
     
     
         29 . The method of  claim 18 , wherein:
 the buffer is 0.1 M HEPES;   the salt is from about 0.25% to about 2% NaCl;   the at least one membrane penetrable sugar is 0.1% w/v to about 12% w/v trehalose;   the neutral dextran with a molecular weight greater than or equal to 50,000 daltons is from about 0.01% w/v to about 25% w/v Dextran-70; and   the glucose is 0.1% w/v to about 12% w/v glucose.   
     
     
         30 . A method of preserving a cancer cell comprising:
 contacting the cancer cell with a solution, wherein the solution comprises:
 0.1 M HEPES; 
 from about 0.25% to about 2% NaCl; 
 0.1%) w/v to about 12%) w/v trehalose; 
 6% w/v Dextran-70; and 
 about 2% w/v glucose; and drying the cell at room temperature.
