Abstract
The present inventors successfully obtained anti-NR10 antibodies having an effective neutralizing activity against NR10. The anti-NR10 antibodies provided by the present invention are useful as, for example, pharmaceuticals for treating or preventing inflammatory diseases.

37 . The pharmaceutical composition of claim 30 , comprising at least one low-molecular-weight polypeptide.
38 . The pharmaceutical composition of claim 30 , comprising serum albumin or gelatin or both.
39 . The pharmaceutical composition of claim 30 , further comprising at least one amino acid.
40 . The pharmaceutical composition of claim 39 , wherein the at least one amino acid is selected from the group consisting of glycine, glutamine, asparagine, arginine, and lysine.
41 . The pharmaceutical composition of claim 30 , wherein the composition is an isotonic aqueous solution suitable for injection.
42 . The pharmaceutical composition of claim 41 , wherein the aqueous solution comprises physiological saline or dextrose or both.
43 . The pharmaceutical composition of claim 41 , comprising an ingredient selected from the group consisting of D-sorbitol, D-mannose, D-mannitol, and sodium chloride.
44 . The pharmaceutical composition of claim 30 , comprising a solubilizing agent.
45 . The pharmaceutical composition of claim 44 , wherein the solubilizing agent is an alcohol, a polyalcohol, or a non-ionic detergent.

55 . A method for producing an anti-NR10 antibody, the method comprising providing a cell containing recombinant nucleic acid encoding an antibody that recognizes domain 1 of NR10 and culturing the cell in vitro so that the antibody is expressed by the cell.

A composition comprising an antibody that recognizes domain 1 of NR10, the antibody being covalently linked to a molecule selected from the group consisting of a polymeric substance, hyaluronic acid, a radioactive substance, a fluorescent substance, a luminescent substance, an enzyme, and a toxin.
57 . A method for obtaining the composition of claim 56 , the method comprising chemically conjugating the molecule to an antibody that recognizes domain 1 of NR10.
58 . A method for measuring the activity of an anti-NR10 antibody, the method comprising contacting the antibody with cells of an interleukin 31 (IL-31)-dependent cell line and determining whether the antibody suppresses growth of the cells.
59 . A method for determining the NR10-binding activity of an anti-NR10 antibody, the method comprising use of a technique selected from ELISA (enzyme-linked immunosorbent assay), EIA (enzyme immunoassay), RIA (radioimmunoassay) and fluorescent antibody assay to determine the NR10-binding activity of the anti-NR10 antibody.
60 . The method of claim 59 , wherein the method comprises use of an enzyme immunoassay that comprises:
(a) contacting a sample containing an anti-NR10 antibody with an NR10-coated plate; (b) adding to the plate a secondary antibody that is labeled with an enzyme, wherein the secondary antibody binds to the anti-NR10 antibody; (c) incubating the plate with the antibodies; (d) washing the plate; (e) adding a substrate for the enzyme to the plate; and (f) measuring absorbance to detect enzymatic alteration of the substrate, as a measure of the NR10-binding activity of the anti-NR10 antibody.
61 . A method for treating or preventing an inflammatory disease in a subject in recognized need thereof, the method comprising administering to the subject an antibody that recognizes domain 1 of NR10.
62 . The method of claim 61 , wherein the inflammatory disease is a disease with pathological features involved in cytological and histological reactions that occur in affected blood vessels and adjacent tissues in response to an injury or abnormal stimulation caused by a physical, chemical, or biological agent.
63 . A method for isolating and purifying an antibody that recognizes domain 1 of NR10, the method comprising subjecting a sample comprising the antibody to a method comprising a combination of multiple techniques selected from column chromatography, filtration, ultrafiltration, salting out, solvent precipitation, solvent extraction, distillation, immunoprecipitation, SDS-polyacrylamide gel electrophoresis, isoelectrofocusing, dialysis, and recrystallization.
64 . The method of claim 63 , wherein the method comprises a column chromatography technique selected from the group consisting of affinity chromatography, ion exchange chromatography, hydrophobic chromatography, gel filtration, reverse phase chromatography, and adsorption chromatography.
65 . The method of claim 64 , wherein the column chromatography technique comprises affinity chromatography using a protein A column or a protein G column.
66 . An anti-NR10 antibody that comprises heavy chain variable region CDRs1-3 comprising SEQ ID NOs: 196, 197, and 184, respectively, and light chain variable region CDRs1-3 comprising SEQ ID NOs: 202, 170, and 193, respectively.
