Abstract

Immunogenic compositions that include swine infertility and respiratory virus (“SRRS virus”) are provided herein. Also provided are kits for producing and using the immunogenic compositions. Vaccines and sera for treatment of SRRS, a method for producing the vaccine, methods for diagnosis of SRRS, and antibodies to the viral agent useful in diagnosis and treatment of SRRS are also provided. Swine infertility and respiratory virus is also referred to herein as Mystery Swine Disease (“MSD”).

1 . A diagnostic kit for detecting a first antibody in a biological sample, wherein the first antibody recognizes an antigen from a swine infertility and respiratory syndrome virus, the kit comprising:
 an antigen that is immunoreactive with serum antibodies of a sow, wherein the serum antibodies are induced by inoculating the sow with an inactivated form of viral agent ATCC VR-2332; and   a label for detecting the first antibody.

         3 . A culture of a fastidious, non-hemagglutinating, enveloped RNA virus, wherein the virus is swine infertility and respiratory syndrome virus ATCC VR-2332. 
     
     
         4 . A immunogenic composition comprising attenuated SIRS virus ATCC VR-2332 which is non-zoopathogenic in swine. 
     
     
         5 . A method for detecting swine infertility and respiratory syndrome virus comprising:
 contacting an antibody and a sample suspected of containing a swine infertility and respiratory syndrome virus to form a complex between the antibody and a viral antigen, wherein the antibody is a monoclonal antibody which specifically binds to a 15 kD antigen of viral agent ATCC VR-2332; and   detecting the complex.   
     
     
         6 . A method of growing swine infertility and respiratory syndrome virus comprising:
 (a) inoculating swine infertility and respiratory syndrome virus on a first culture of simian cells;   (b) incubating the inoculated first culture;   (c) lysing the incubated first culture prior to the appearance of cytopathic effects in the first culture to form a lysate;   (d) inoculating the lysate on a second culture of simian cells;   (e) incubating the inoculated second culture; and   (f) titrating the swine infertility and respiratory syndrome virus comprising:
 (i) inoculating serial dilutions of the swine infertility and respiratory syndrome virus on simian cells; 
 (ii) incubating the inoculated simian cells for at least 5 days; and 
 (iii) calculating the 50% tissue culture infective dose (TCID 50 ) of the swine infertility and respiratory syndrome virus. 
   
     
     
         7 . The method of  claim 6 , wherein trypsin is added to at least one of the first and second inoculated culture. 
     
     
         8 . The method of  claim 6 , wherein the simian cells are MA-104 cells. 
     
     
         9 . The method of  claim 6 , wherein the swine infertility and respiratory syndrome virus is part of a swine tissue homogenate. 
     
     
         10 . The method of  claim 6 , wherein the swine infertility and respiratory syndrome virus is derived from a swine tissue homogenate.
