Abstract
The present invention relates to engineered multivalent and multispecific binding proteins, methods of making, and specifically to their uses in the prevention, diagnosis, and/or treatment of disease.

77 . A binding protein comprising a polypeptide chain, wherein said polypeptide chain comprises VD1-(X1)n-VD2-C-(X2)n, wherein;
VD1 is a first heavy chain variable domain; VD2 is a second heavy chain variable domain; C is a heavy chain constant domain; X1 is a linker; X2 is an Fc region; and wherein the binding protein is capable of binding a pair of antigens selected from the group consisting of TNF and IL-17A; TNF and RANKL; TNF and VEGF; TNF and SOST; TNF and DKK; TNF and alphaVbeta3; TNF and NGF; TNF and IL-23p19; TNF and IL-6; TNF and IL-6R; TNF and CD-20; IgE and IL-13; IL-13 and IL-23p19; IgE and IL-4; IgE and IL-9; IL-13 and IL-9; IL-13 and IL-4; IL-6R and VEGF; IL-6R and IL-17A; IL-6R and RANKL; IL-17A and IL-I beta; IL-I beta and RANKL; IL-L beta and VEGF; and RANKL and CD-20.

The binding protein according to claim 77 , wherein VD1 and VD2 heavy chain each independently comprise an amino acid sequence selected from the group consisting of SEQ ID NOs: 28, 30, 32, 34, 36, 38, 40, 42, 44, 46, 48, 50, 54, 56, 58, 60, 62, 64, 66, and 70 and wherein the VD1 and VD2 light chain variable domains each independently comprise an amino acid sequence selected from the group of consisting of SEQ ID NOs: 29, 31, 33, 35, 37, 39, 41, 43, 45, 47, 49, 51, 55, 57, 59, 61, 63, 65, 67, and 71.
79 . The binding protein according to claim 77 , wherein X1 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs 1-27.
80 . The binding protein according to claim 77 , wherein the binding protein comprises two first polypeptide chains and two second polypeptide chains and four functional binding sites.
81 . The binding protein according to claim 77 , wherein the Fc region is a native sequence Fc region or variant sequence Fc region.
82 . The binding protein according to claim 81 , wherein the Fc region is an IgG1, IgG2, IgG3, IgG4, IgA, IgM, IgE, or IgD.

The binding protein according to claim 77 , wherein said binding protein has an on rate constant (Kon) to said one or more targets selected from the group consisting of: at least about 10 2 M −1 s −1 ; at least about 10 3 M −1 s −1 ; at least about 10 4 M −1 s −1 ; and at least about 10 6 M −1 s −1 and as measured by surface plasmon resonance.

The binding protein according to claim 77 , wherein said binding protein has an off rate constant (Koff) to said one or more targets selected from the group consisting of: at most about 10 −3 s −1 ; at most about 10 4 s −1 ; at most about 10 −5 s −1 ; and at most about 10 −6 s −1 as measured by surface plasmon resonance.

The binding protein according to claim 77 , wherein said binding protein has a dissociation constant (K D ) to said one or more targets selected from the group consisting of: at most about 10 −7 M; at most about 10 −8 M; at most about 10 −9 M; at most about 10 −10 M; at most about 10 −11 M; at most about 10 −12 M; and at most 10 −13 M.
86 . A binding protein conjugate comprising the binding protein according to claim 77 , said binding protein conjugate further comprising an agent selected from the group consisting of an immunoadhension molecule, an imaging agent, a therapeutic agent, and a cytotoxic agent.
87 . The binding protein conjugate according to claim 86 , wherein said agent is an imaging agent selected from the group consisting of a radiolabel, an enzyme, a fluorescent label, a luminescent label, a bioluminescent label, a magnetic label, and biotin.
88 . The binding protein conjugate according to claim 87 , wherein said imaging agent is a radiolabel selected from the group consisting of 3 H, 14 C, 35 S, 90 Y, 99 Tc, 111 In, 125 I, 131 I, 177 Lu, 166 Ho and 153 Sm.

The binding protein conjugate according to claim 86 , wherein said agent is a therapeutic or cytotoxic agent selected from the group consisting of an anti-metabolite, an alkylating agent, an antibiotic, a growth factor, a cytokine, an anti-angiogenic agent, an anti-mitotic agent, an anthracycline, toxin, and an apoptotic agent.
90 . The binding protein according to claim 77 , wherein said binding protein is a crystallized binding protein.
91 . The binding protein according to claim 90 , wherein said crystal is a carrier-free pharmaceutical controlled release crystal.
92 . The binding protein according to claim 90 , wherein said binding protein has a greater half-life in vivo than the soluble counterpart of said binding protein.
93 . The binding protein according to claim 90 , wherein said binding protein retains biological activity.
94 . An isolated nucleic acid encoding a binding protein amino acid sequence according to claim 77 .
95 . A vector comprising an isolated nucleic acid according to claim 94 .
96 . The vector according to claim 95 , wherein said vector is selected from the group consisting of pcDNA, pTT, pTT3, pEFBOS, pBV, pJV, pcDNA3.1 TOPO, pEF6, pHybE, TOPO, and pBJ.
97 . A host cell comprising a vector according to claim 95 .
98 . The host cell according to claim 97 , wherein said host cell is a prokaryotic cell.

100 . A method of producing a binding protein, comprising culturing the host cell of claim 97 in culture medium under conditions sufficient to produce the binding protein.
101 . A pharmaceutical composition comprising the binding protein of claim 77 , and a pharmaceutically acceptable carrier.
102 . The pharmaceutical composition of claim 101 , further comprising at least one additional therapeutic agent.
