Abstract

Disclosed herein, in certain embodiments, is an HC•HA complex comprising hyaluronan and a heavy chain of IαI, wherein the transfer of the heavy chain of IαI is catalyzed by TSG-6. Further disclosed herein, in certain embodiments, is an HC•HA complex comprising hyaluronan and a heavy chain of IαI, wherein the transfer of the heavy chain of IαI is catalyzed by the TSG-6 like protein. Additionally, disclosed herein are methods of manufacturing said complex and methods of use thereof.

What is claimed is: 
     
         1 . A pharmaceutical composition, comprising
 a) reconstituted HC•HA (rcHC•HA) complex comprising hyaluronan (HA) and a heavy chain of IαI, wherein TSG-6 is not covalently associated with the complex; and   b) a pharmaceutically-acceptable excipient.   
     
     
         2 . The rcHC•HA of  claim 1 , wherein the heavy chain of IαI is HC1, HC2, or a combination thereof. 
     
     
         3 . The rcHC•HA of  claim 2 , wherein HC1 and HC2 of IαI are recombinant. 
     
     
         4 . The composition of  claim 1 , further comprising HC•HA complex isolated from amniotic material complex (nHC•HA). 
     
     
         5 . The composition of  claim 1 , wherein the nHC•HA complex is isolated from chorionic membrane, amniotic membrane, or a combination thereof. 
     
     
         6 . The composition of  claim 1 , further comprising: an anti-inflammatory agent, an anti-scarring agent, a chemotherapeutic agent, an immuno-suppressive agent, or a combination thereof. 
     
     
         7 . An HC•HA (rcHC•HA) complex intermediate, comprising hyaluronan (HA) and a heavy chain of IαI, wherein HA is immobilized on a stationary support via HA Binding Protein (HABP) crosslinked to the stationary support. 
     
     
         8 . The rcHC•HA of  claim 7 , wherein the heavy chain of IαI is HC1, HC2, or a combination thereof. 
     
     
         9 . The rcHC•HA of  claim 8 , wherein HC1 and HC2 of IαI are recombinant. 
     
     
         10 . The rcHC•HA complex of  claim 8 , wherein HC1 and HC2 of IαI are purified from serum. 
     
     
         11 . The rcHC•HA complex of  claim 8 , wherein HC1 and HC2 of IαI are from unpurified serum. 
     
     
         12 . The rcHC•HA of  claim 8 , wherein at least one of HA, HC1, and HC2 is generated by a plurality of cells.
