Abstract
Methods and compositions are provided for the identification and isolation of mammalian HLA-G + MSC, HLA-E + MSC, or HLA-G + /HLA-E + MSC. The methods of the invention provide a means to obtain enriched HLA-G + MSC, HLA-E + MSC, or HLA-G + /HLA-E + MSC populations.

1 . A substantially homogeneous cellular composition of mammalian mesenchymal stem cells (MSC) comprising an enriched population of HLA-G+ MSC, HLA-E+ MSC, and/or HLA-G+/HLA-E+ MSC.
2 . The composition of claim 1 , wherein at least about 25% of the cells present in said composition are HLA-G+ MSC.
3 . The composition according to claim 2 , wherein said cells that have an HLA-G+ MSC phenotype are present after initial isolation from tissue.
4 . The composition of claim 1 , wherein at least about 25% of the cells present in said composition are HLA-E+ MSC.
5 . The composition of claim 1 , wherein at least about 25% of the cells present in said composition are HLA-G+/HLA-E+ MSC.
6 . The composition according to claim 1 , wherein said MSC are isolated from adipose tissue.
7 . The composition according to claim 1 , wherein said MSC are isolated from bone marrow.
8 . The composition according to claim 1 , wherein at least one MSC of said population further expresses indoleamine-pyrrole 2,3, dioxygenase (INDO).
9 . The composition according any one of claims 1 and 8 , wherein at least one MSC of said population further expresses CD200.
10 . The composition according to claim 1 , wherein at least one MSC of said composition comprises an exogenous nucleic acid.
11 . The composition according to claim 10 , wherein said exogenous nucleic acid comprises a coding sequence for a polypeptide sequence of interest, operably connected to a promoter sequence active in said HLA-G+ MSC.
12 . The composition according to claim 11 , wherein said exogenous nucleic acid comprises a sequence encoding HLA-G.
13 . A method for isolating mesenchymal stem cells (MSC), the method comprising:
contacting a population of MSC with an antibody specific for HLA-G and/or HLA-E for a period of time sufficient to bind said HLA-G and/or HLA-E; selecting for cells expressing said HLA-G and/or HLA-E.
14 . The method according to claim 13 , wherein said population of cells are isolated from adipose tissue.
15 . The method according to claim 13 , wherein said population of cells are isolated from bone marrow.Cited by (0)
